規(guī);N豬場(chǎng)豬藍(lán)耳病和豬瘟抗體變化規(guī)律研究及在健康流程管理中的應(yīng)用

【摘要】:豬繁殖與呼吸綜合征(PRRS),又稱豬藍(lán)耳病,是由豬繁殖與呼吸綜合征病毒引起的一種以母豬繁殖障礙和仔豬呼吸道疾病為特征的烈性傳染病,該病已在全世界范圍內(nèi)廣泛傳播,嚴(yán)重影響?zhàn)B豬業(yè)的發(fā)展,被世界衛(wèi)生組織列為需要通報(bào)的傳染病之一。豬瘟(CSF)是由豬瘟病毒引起的一種豬的高度接觸性傳染病,具有高度接觸傳染性,流行廣泛,發(fā)病率、死亡率高,危害極大;是經(jīng)濟(jì)損失最為嚴(yán)重的傳染病之一,也被世界衛(wèi)生組織列為需要通報(bào)的傳染病之一。高致病性豬藍(lán)耳病與豬瘟均是國(guó)家中長(zhǎng)期動(dòng)物疫病防治規(guī)劃(2012-2020年)優(yōu)先防治的國(guó)內(nèi)16種動(dòng)物疫病中的二類動(dòng)物疫病。豬瘟,特別是豬繁殖與呼吸綜合征是免疫抑制性疾病,在豬場(chǎng)養(yǎng)殖生產(chǎn)中難以凈化、進(jìn)行有效地預(yù)防控制,防控效果不理想等,對(duì)養(yǎng)豬業(yè)的健康發(fā)展帶來較大的阻礙。因此,本研究從群體免疫學(xué)著手,結(jié)合豬場(chǎng)生產(chǎn)成績(jī),開展了規(guī);N豬場(chǎng)豬藍(lán)耳病和豬瘟抗體變化規(guī)律研究,總結(jié)確定出豬場(chǎng)穩(wěn)定生產(chǎn)時(shí)PRRS和CSF的基準(zhǔn)線的標(biāo)準(zhǔn)區(qū)間(范圍),以此分析判定疫病發(fā)生風(fēng)險(xiǎn),調(diào)整免疫方案與飼養(yǎng)管理,并作為豬場(chǎng)健康流程管理中動(dòng)態(tài)篩選和重點(diǎn)管理的判定依據(jù)之一,通過管理應(yīng)用實(shí)例,為豬場(chǎng)提供一個(gè)養(yǎng)殖與疫病防控相結(jié)合的管理模式,同時(shí)也為豬藍(lán)耳病和豬瘟控制計(jì)劃提供技術(shù)支撐。建立規(guī);N豬場(chǎng)豬繁殖與呼吸綜合征血清學(xué)抗體評(píng)價(jià)基準(zhǔn)線區(qū)間。通過2010-2013年時(shí)間開展規(guī);N豬場(chǎng)豬藍(lán)耳病抗體水平的監(jiān)測(cè),掌握豬群藍(lán)耳病抗體水平S/P值、離散度的變化規(guī)律,結(jié)合豬場(chǎng)生產(chǎn)成績(jī),確定判定豬場(chǎng)PRRS免疫后豬群處于穩(wěn)定狀態(tài)時(shí)藍(lán)耳病抗體水平以及整齊度范圍,從多個(gè)規(guī);i場(chǎng)PRRS抗體變化數(shù)據(jù)的跟蹤分析結(jié)果,結(jié)合豬場(chǎng)實(shí)際生產(chǎn)情況與業(yè)績(jī),總結(jié)出,具有較好生產(chǎn)成績(jī)的豬場(chǎng),其PRRS抗體基準(zhǔn)線區(qū)間確定平均值在1.0~2.0區(qū)間,離散度分布在50~70區(qū)間。以此作為參考評(píng)估藍(lán)耳病疫苗免疫效果的指標(biāo)之一,及時(shí)做好免疫程序的調(diào)整,加強(qiáng)流程管理,從而降低豬群藍(lán)耳病的發(fā)病幾率甚至做出預(yù)警,為PRRS的預(yù)防和控制提供臨床參考資料。建立規(guī);N豬場(chǎng)豬瘟血清學(xué)抗體評(píng)價(jià)基準(zhǔn)線區(qū)間與范圍,開展了豬瘟疫病凈控制管理研究。選取2個(gè)生產(chǎn)業(yè)績(jī)良好的規(guī)模化種豬場(chǎng),進(jìn)行2011-2014年期間豬瘟抗體監(jiān)測(cè)數(shù)據(jù)分析變化規(guī)律,結(jié)合豬場(chǎng)實(shí)際生產(chǎn)數(shù)據(jù),確定豬瘟疫苗免疫效果良好時(shí),其抗體陽(yáng)性阻斷率、離散度和抗體平均值標(biāo)準(zhǔn)區(qū)間,免疫豬瘟疫苗后其抗體平均阻斷率應(yīng)達(dá)到50以上,離散度CV值應(yīng)在25以下,可視為免疫效果良好。當(dāng)有效免疫率達(dá)85%以上發(fā)生豬瘟機(jī)率較小,而小于50%則顯示免疫無效或?yàn)椴环(wěn)定群體,應(yīng)進(jìn)一步加強(qiáng)免疫。以此評(píng)價(jià)豬瘟免疫效果,調(diào)整免疫方案與管理重點(diǎn),降低豬群豬瘟的發(fā)病幾率。為豬瘟的預(yù)防和控制提供臨床參考資料。通過建立的規(guī);N豬場(chǎng)自身的PRRS和CSF抗體基準(zhǔn)線區(qū)間(范圍),開展其在豬場(chǎng)健康流程管理中的應(yīng)用,并結(jié)合飼養(yǎng)管理方案,做好方案實(shí)施前后的比較分析,總結(jié)歸納出健康管理的方案與流程。研究表明,規(guī);N豬場(chǎng)的管理應(yīng)當(dāng)根據(jù)不同階段進(jìn)行分點(diǎn)式管理,把握不同環(huán)節(jié)飼養(yǎng)管理重點(diǎn),借助日常管理數(shù)據(jù)和疫病抗體水平規(guī)律,探索出豬場(chǎng)豬藍(lán)耳病、豬瘟等重要疫病抗體水平標(biāo)準(zhǔn)區(qū)間,加之動(dòng)態(tài)管理,減少疫病發(fā)生風(fēng)險(xiǎn),以此做好種豬場(chǎng)疫病的凈化與控制計(jì)劃,達(dá)到豬場(chǎng)的穩(wěn)定生產(chǎn)。健康管理的總體思路主要以生物安全、繁育體系、流程控制、群體健康、疫苗免疫為主,配套規(guī)程管理、動(dòng)態(tài)監(jiān)測(cè)、優(yōu)勝劣汰。目前,健康流程管理已作為國(guó)外很多規(guī);i場(chǎng)的日常管理理念,通過日常監(jiān)測(cè)與階段管理,不僅是豬場(chǎng)穩(wěn)定生產(chǎn)的重要因素,也是豬瘟等重要疫病凈化與控制工作實(shí)施成功的手段之一,本研究工作為規(guī);i場(chǎng)的健康管理提供了一個(gè)技術(shù)示范與臨床成功實(shí)例。針對(duì)目前高致病性藍(lán)耳病變異株及疫苗毒株的多種性,而目前的檢測(cè)技術(shù)難以判定區(qū)分PRRS疫苗株和野毒株的現(xiàn)狀。建立了高分辨率溶解曲線HRM分析技術(shù)平臺(tái),研究出區(qū)別PRRS經(jīng)典株和變異株的PCR-HRM方法,區(qū)分高致病性豬繁殖與呼吸綜合征活疫苗GDr180株、TJM-F92株與野毒株的PCR-HRM檢測(cè)方法;區(qū)分高致病性豬繁殖與呼吸綜合征JXA1-R疫苗毒株和野毒株的PCR-HRM檢測(cè)方法。為豬繁殖與呼吸綜合征鑒別診斷提供技術(shù)支撐,解決了生產(chǎn)實(shí)際需求。
[Abstract]:Porcine reproductive and respiratory syndrome (PRRS), * * *, is a porcine reproductive and respiratory syndrome virus caused by porcine reproductive and respiratory syndrome virus, a sow reproductive disorder and piglets respiratory diseases characterized by strong infectious diseases, the disease has been widely spread throughout the world, seriously affecting the development of pig industry, WHO is listed as a notification. CSF is a highly contagious infectious disease caused by hog cholera virus. It is highly contagious and widespread, with high morbidity, high mortality and great harm. It is one of the most serious infectious diseases, and is also listed as one of the infectious diseases that need to be notified by WHO. Classical swine fever is the two type of animal disease that is the first priority in the national middle and long term animal disease prevention program (2012-2020 *) to control 16 domestic animal diseases. Swine fever, especially porcine reproductive and respiratory syndrome, is an immunosuppressive disease. It is difficult to purify in pig breeding, effective prevention and control, and poor control and prevention. Therefore, this study started with group immunology and combined with pig farm production results, and carried out a study on the variation pattern of pig ear * s disease and swine fever antibody in large scale pig farms. The standard inter area (range) of PRRS and CSF for stable production in pig farms was determined. Integrated immunization and feeding management, as one of the criteria for dynamic screening and key management of pig farms health process management, provides a management mode for pig farms to combine culture with disease prevention and control through management application examples, and also provides technical support for * pig ear disease and swine fever control plan. * The baseline intervals of serological antibodies for reproductive and respiratory syndrome were evaluated. The antibody level of pig * s disease in large scale pig farms was monitored for 2010-2013 years. The S/P level of the antibody level and the variation rule of the dispersion of pigs were mastered. Combined with the production performance of pig farms, the blue ear was determined when the pigs were in stable state after PRRS vaccination. Based on the results of tracking analysis of PRRS antibody change data from several large-scale pig farms, combined with the actual production situation and performance of pig farms, it is concluded that the PRRS antibody baseline interval of pig farms with good production performance is between 1.0 and 2.0, and the dispersion is between 50 and 70. One of the indicators for evaluating the immune effect of blue-ear disease vaccine is to adjust the immune procedure in time and strengthen the process management so as to reduce the incidence of blue-ear disease in pigs and even make early warning, providing clinical reference for the prevention and control of PRRS. * study on the net control and management of swine plague. Choose 2 large scale pig farms with good production performance, analyze the variation rules of antibody against swine fever during 2011-2014 years, and combine the actual production data of pig farms to determine the positive blocking rate, dispersion and standard interval of antibody against swine fever vaccine. After vaccination, the average blocking rate of antibody should be above 50, and the CV value of dispersion should be below 25, which can be regarded as a good immune effect. When the effective immune rate is above 85%, the incidence of swine fever is small, and less than 50% shows that the immune response is ineffective or unstable, the immunization should be further strengthened. To reduce the incidence of swine fever and provide clinical reference for the prevention and control of swine fever, the application of PRRS and CSF antibody baseline in the health process management of swine farms was carried out by establishing PRRS and CSF antibody baseline intervals (ranges) of large-scale breeding pig farms. Conclusion: the management of large scale pig farms should be divided into different stages according to different stages, and the key points of different links should be grasped. With the help of daily management data and epidemic disease antibody levels, the standard interval of antibody levels for pig diseases such as pig ear disease *, swine fever and other important diseases should be explored. The general idea of health management is mainly biological safety, breeding system, process control, population health, vaccine immunization, supporting procedures management, dynamic monitoring, survival of the fittest. As a daily management concept of many large-scale pig farms abroad, daily monitoring and stage management is not only an important factor for stable production of pig farms, but also one of the successful means for the purification and control of important epidemic diseases such as swine fever. This study provides a technical demonstration and clinical success for the health management of large-scale pig farms. For example, in view of the diversity of the highly pathogenic RT variants and vaccine strains, the current detection technology is difficult to determine the status of PRRS vaccine strains and wild strains.
