內(nèi)源干擾物和中藥小分子對斑馬魚生長發(fā)育的影響

【摘要】:內(nèi)源干擾物對生長發(fā)育的影響還沒有得到充分的研究,然而早期接觸內(nèi)源干擾物可能會在之后的發(fā)育中產(chǎn)生永久有害的影響。轉(zhuǎn)基因斑馬魚的組織由于結(jié)合了熒光蛋白而成為檢測給定物質(zhì)有無影響的強大工具。我們利用7種轉(zhuǎn)基因株系檢測了6種己知的內(nèi)源干擾物和3種藥物對于斑馬魚生長發(fā)育的影響。檢測表明其中4種物質(zhì)對于斑馬魚的四個不同器官組織生長發(fā)育產(chǎn)生了影響。內(nèi)源干擾物四溴雙酚A和3種種藥物擾亂了斑馬魚血管系統(tǒng)的正常發(fā)育。其中組織蛋白去乙酰酶抑制劑曲古菌素A和丙戊酸還抑制了斑馬魚內(nèi)分泌胰腺,外分泌胰腺,肝臟和咽頭齒的生長發(fā)育。實驗結(jié)果表明內(nèi)源干擾物影響了斑馬魚的生長發(fā)育。斑馬魚轉(zhuǎn)基因株系是活體快速檢測內(nèi)源干擾物對胚胎生長發(fā)育影響的良好模型。傳統(tǒng)中藥是現(xiàn)代中藥的重要組成部分。然而對于傳統(tǒng)中藥及其有效成分對于成長發(fā)育影響的研究還比較少。我們利用斑馬魚對三種常見中藥及其活性成分進行了測試。研究表明木通和黃芪脫水溶液對延遲了斑馬魚胚胎的生長發(fā)育,丹參脫水溶液及其三種主要活性物質(zhì)單獨或混合溶液對斑馬魚的血管系統(tǒng)的生長發(fā)育有不同程度的抑制作用。研究結(jié)果表明斑馬魚是進行小物質(zhì)篩選的良好模型。實驗結(jié)果也我們將內(nèi)源干擾物和中藥活性成為聯(lián)系在一起,比如他們可能會有相似的作用目標,核受體。
[Abstract]:The effects of endogenous interference on growth and development have not been fully studied, however, early exposure to endogenous interference may have a permanent harmful effect in subsequent development. Genetically modified zebrafish tissues are powerful tools for detecting the effects of a given substance by combining fluorescent proteins. The effects of 6 known endogenous disruptors and 3 drugs on the growth and development of zebrafish were studied using 7 transgenic lines. The results showed that four of them affected the growth and development of four different organs of zebrafish. Intrinsically, tetrabromobisphenol A and 3 drugs disrupt the normal development of the zebrafish vascular system. Among them, histone deacetylase inhibitor Tracogenin A and valproic acid also inhibited the growth and development of the endocrine pancreas, exocrine pancreas, liver and pharyngeal head teeth of zebrafish. The results showed that the endogenous interference affected the growth and development of zebrafish. Zebrafish transgenic lines are a good model for rapid detection of the effects of endogenous interference on embryo growth and development in vivo. Traditional Chinese medicine is an important part of modern Chinese medicine. However, there are few studies on the effects of traditional Chinese medicine and its active components on growth and development. Three common Chinese medicines and their active components were tested by zebrafish. The results showed that Astragalus and Astragali dehydrated solution delayed the growth and development of zebrafish embryos, and the dehydration solution of Salvia miltiorrhiza and its three main active substances alone or mixed had different inhibitory effects on the growth and development of the vascular system of zebrafish. The results show that zebrafish is a good model for screening small substances. The results also linked endogenous disruptions to the activity of traditional Chinese medicine, for example, they might have similar targets, nuclear receptors.
