【摘要】:本論文對蘚葉卷瓣蘭(Bulbophyllum retusiusculum)、芳香石豆蘭(Bulbophyllum ambrosia)和韭菜(Allium tuberosum)根的化學(xué)成分進(jìn)行了研究。實(shí)驗(yàn)中應(yīng)用硅膠柱層析,反向(RP-18)柱層析,葡聚糖凝膠(Sephadex LH-20)柱層析,大孔樹脂HPD101柱層析等多種分離材料和分離技術(shù),同時應(yīng)用波譜分析方法和化學(xué)方法,從以上3種植物中分離鑒定了93個化合物,其中新化合物17個。論文還對分離得到的部分化合物進(jìn)行了活性篩選,發(fā)現(xiàn)了一些有明顯活性的化合物。蘚葉卷瓣蘭(Bulbophyllum retusiusculum)是蘭科(Orchidaceae)石豆蘭屬(Bulbophyllum)植物。目前對蘚葉卷瓣蘭(Bulbophyllum retusiusculum)的化學(xué)成分研究還未見報道。本論文對蘚葉卷瓣蘭的化學(xué)成分進(jìn)行了深入研究,從中分離得到37個化合物,包括9個新化合物:retusiusines A-G (1-7), bulbotetusine (8), apigenin 6-C-α-arabino-furanosyl 8-C-a-arabinopyranoside (9)和28個已知化合物。主要化學(xué)成分為苯丙素類化合物、聯(lián)芐類化合物、菲類和二氫菲類化合物。對9個新化合物進(jìn)行了細(xì)胞毒活性篩選實(shí)驗(yàn),結(jié)果表明化合物4,5和7具有較強(qiáng)的抗肝癌(SMMC-7721)活性,化合物4和5有較弱的抗白血病(HL-60)活性,化合物7具有一定的抗乳腺癌(MCF-7)活性。另外還對化合物1,2,4,9,14,21,22,24和26進(jìn)行了抗菌活性篩選,結(jié)果顯示對于白色念珠菌(C. albicans),化合物2有較強(qiáng)的抗菌活性,化合物22有中等強(qiáng)度的抗菌活性,化合物4,21,24和26也有微弱的抗菌活性;對枯草芽孢桿菌(B. subtilis),化合物21,22,24和26有微弱的抗菌活性。芳香石豆蘭(Bulbophyllum ambrosia),是蘭科(Orchidaceae)石豆蘭屬(Bulbophyllum)植物。具有清熱,止咳的作用,主要用于治療肺熱咳嗽。目前對芳香石豆蘭的化學(xué)成分研究還未見報道,本論文對芳香石豆蘭的化學(xué)成分進(jìn)行了深入分析。從中分離鑒定了30個化合物,包括1個新化合物amrosine (1)和29個已知化合物。該植物主要化學(xué)成分為苯丙素類化合物、聯(lián)芐類化合物、菲類和二氫菲類化合物。韭(Allium tuberosum)為百合科蔥屬多年生草本植物,其莖、葉、花均可食用,是云南日常食用的蔬菜,也可用于治療腹痛、腹瀉、吐血、毒蛇咬傷、哮喘等。目前研究主要集中在韭菜籽,而對于韭菜根的研究較少,為了對韭菜進(jìn)行深入的開發(fā)利用,本文對韭菜根的化學(xué)成分進(jìn)行了研究。從中分離得到了26個化合物,其中有7個新化合物tuberosines A-G(1-7)和19個已知化合物。7個新化合物包括3個甾體皂苷化合物(1-3),1個苯丙酸甲酯苷類化合物(4),3個苯乙酮苷類化合物(5-7)。還對分離到的3個新的甾體皂苷化合物(1-3)進(jìn)行了抗腫瘤活性研究,結(jié)果表明化合物3對肝癌(SMMC-7721)、肺癌(A-549)、結(jié)腸癌(SW480)、白血病(HL-60)和乳腺癌(MCF-7)均具有活性。對分離到的9個化合物(化合物1-5,8,18,23和24)進(jìn)行了抗菌活性篩選,結(jié)果顯示化合物23和24對枯草桿菌(B. subtilis,32 μg/mL)和大腸桿菌(E. coil,16 μg/mL)有一定的抗菌活性,化合物2對枯草桿菌(B. subtilis, 64 μg/mL)和大腸桿菌(E. coil,64 μg/mL)也有一定的抗菌活性,化合物3和18對枯草桿菌(B. subtilis,128μg/mL)有微弱的抗菌活性,其它化合物沒有抗菌活性。本論文最后對2015年及以前發(fā)表的石豆蘭屬(Bulbophyllum)植物化學(xué)成分及藥理活性研究進(jìn)行了綜述,共涉及105個化合物和31篇文獻(xiàn)。
[Abstract]:The moss leaf cactus (Bulbophyllum retusiusculum), fragrant orchid (Bulbophyllum Ambrosia) and Shi Doulan (Allium tuberosum) the chemical composition of leek root was investigated by silica gel column chromatography experiment, reverse (RP-18) column chromatography, Sephadex column chromatography (Sephadex LH-20), HPD101 macroporous resin column chromatography and other separation materials and separation technology, at the same time the application of spectral analysis and chemical methods, from more than 3 kinds of plants in the isolation of 93 compounds, including 17 new compounds. Shi Doulan (Bulbophyllum Ambrosia), fragrant orchid (Orchidaceae) is a species of Bulbophyllum (Bulbophyllum) plant. With heat, cough, mainly used for the treatment of lung heat cough. At present the studies on chemical constituents of fragrant Bulbophyllum have been reported, the chemical composition of aromatic Bulbophyllum odoratissimum were analyzed. We obtained 26 compounds isolated from, including 7 new compounds tuberosines A-G (1-7) and 19 known compounds.7 new compounds including 3 steroidal saponins compounds (1-3), 1 benzene methyl propionate glycosides (4, 3) acetophenone glycosides (5-7). 3 new steroidal saponins compounds on isolated (1-3) studied the antitumor activity, the results showed that the compound 3 on hepatocellular carcinoma (SMMC-7721), lung cancer (A-549), colon cancer (SW480), leukemia (HL-60) and breast cancer (MCF-7) were activated. 9 compounds isolated (compounds 1-5,8,18,23 and 24) were screened for antibacterial activity. The results showed that. 23 and 24 of Bacillus subtilis (B. subtilis, 32 g/mL) and Escherichia coli (E. coil, 16 g/mL) has certain antibacterial activity, 2 compounds of Bacillus subtilis (B. subtilis, 64 g/mL) and Escherichia coli (E. coil, 64 g/mL) also has certain antibacterial activity, compounds 3 and 18 of Bacillus subtilis (B. subtilis, 128 g/mL) has weak antibacterial activity, other compounds have no antibacterial activity. In the end of 2015 and previously published species of Bulbophyllum (Bulbophyllum) study on chemical constituents and pharmacological activities of plants were reviewed, involving a total of 105 compounds and 31 articles.
