酸降解法制備褐藻寡糖抗氧化性的研究

【摘要】:從褐藻膠中提取出聚甘露糖醛酸和聚古羅糖醛酸,利用鹽酸降解聚甘露糖醛酸和聚古羅糖醛酸1、2、6 h分別制備了甘露糖醛酸組分(M_1、M_2和M_3)以及古羅糖醛酸組分(G_1、G_2和G_3),并且以肌肽和甘露糖為對照評估寡糖對DPPH自由基、超氧自由基和羥自由基的清除作用。結(jié)果表明,甘露糖醛酸以及古羅糖醛酸對DPPH自由基、超氧自由基具有良好的清除作用,且清除作用隨著寡糖中還原糖含量的增加而增加。在DPPH體系中,M_3的清除效果要好于肌肽,G_3的清除效果與肌肽相近。在超氧自由基體系中,M_3的清除作用高于M_2和M_1,而G_3的清除作用略低于肌肽。在羥自由基體系中,甘露糖醛酸和古羅糖醛酸的清除效果低于肌肽和甘露醇。實驗表明,酸法制備的褐藻寡糖具有較強的抗氧化能力,且抗氧化效果與寡糖的平均聚合度含量有關(guān)。
[Abstract]:Poly mannouronic acid and polyguuronic acid were extracted from alginate. The mannouronic acid fractions, M1M2 and M3, were prepared by hydrochloric acid degradation of polymannouronic acid and polyguuronic acid for 6 h, respectively. The G1G2 and G3S fractions of galuronic acid were prepared, respectively. The DPPH free radicals were evaluated by carnosine and mannose. Scavenging effect of superoxide radical and hydroxyl radical. The results showed that mannouronic acid and uronic acid had a good scavenging effect on DPPH free radical and superoxide free radical, and the scavenging effect increased with the increase of reducing sugar content in oligosaccharides. In DPPH system, the scavenging effect of M _ 3 was better than that of carnosine G3 and the effect was similar to that of carnosine. In the superoxide free radical system, the scavenging effect of M _ S _ 3 is higher than that of M _ S _ 2 and M _ 1, but the scavenging effect of GSP _ 3 is slightly lower than that of carnosine. In hydroxyl radical system, the scavenging effect of mannouronic acid and uronic acid was lower than that of carnosine and mannitol. The results showed that the alginate oligosaccharides prepared by acid method had strong antioxidant capacity and the antioxidant effect was related to the average degree of polymerization of oligosaccharides.
