沙棘水溶性膳食纖維固體發(fā)酵法制備的研究

【摘要】:水溶性膳食纖維(soluble dietary fiber,SDF)是指不能被人體消化道分泌的消化酶所消化,但可溶于熱水,且其水溶液能被四倍乙醇沉淀的那部分膳食纖維。水溶性膳食纖維以其重要的生理功能,越來(lái)越被國(guó)內(nèi)外學(xué)者及民眾所認(rèn)識(shí)和關(guān)注。本實(shí)驗(yàn)以里氏木霉發(fā)酵法發(fā)酵沙棘粕,提高沙棘水溶性膳食纖維的提取率,并對(duì)發(fā)酵后提取的沙棘水溶性膳食纖維進(jìn)行脫色處理,并對(duì)獲得的沙棘水溶性膳食纖維的特性進(jìn)行了測(cè)定與分析。 本試驗(yàn)通過(guò)對(duì)三種發(fā)酵菌株的篩選,發(fā)現(xiàn)里氏木霉菌株適合作為沙棘粕固體發(fā)酵菌株。通過(guò)單因素試驗(yàn)和正交試驗(yàn)可知,影響發(fā)酵法制備沙棘水溶性膳食纖維的因素大小依次為接菌量發(fā)酵時(shí)間發(fā)酵溫度水分含量。最佳工藝條件為:接種量5 mL·100 mg~(-1),發(fā)酵溫度28℃,發(fā)酵時(shí)間6 d,水分加入量2 mL·g~(-1),沙棘水溶性膳食纖維的提取率可達(dá)9.65 %。 通過(guò)對(duì)脫色劑的選擇試驗(yàn),發(fā)現(xiàn)粉末狀活性炭適合作為發(fā)酵制備的沙棘水溶性膳食纖維的脫色。單因素試驗(yàn)和正交試驗(yàn)可知,沙棘水溶性膳食纖維脫色最佳工藝條件為:脫色液pH值為5,活性炭加入量0.5 %,脫色溫度為40℃。脫色后的沙棘水溶性膳食纖維為淡黃色。 通過(guò)對(duì)沙棘水溶性膳食纖維特性研究發(fā)現(xiàn),沙棘水溶性膳食纖維為典型的多糖類物質(zhì),單糖的摩爾比為,鼠李糖:阿拉伯糖:木糖:甘露糖:葡萄糖:半乳糖= 2.93:4.06:4.58:6.13:6.63:28.06。發(fā)酵作用能提高沙棘水溶性膳食纖維的持水力(6.4 g/g)和膨脹力(5.3 mL/g)。沙棘SDF溶液粘度研究發(fā)現(xiàn),隨著沙棘SDF濃度的增加粘度直線增大,隨溫度的升高降低,且受pH值影響較大。此外,沙棘SDF溶液粘度受食鹽濃度影響較小,但受蔗糖濃度的影響較大。沙棘SDF對(duì)O~(2-)和·OH具有一定的清除作用,對(duì)DPPH(1,1-二苯基-2-三硝基苯肼)清除作用較小。
[Abstract]:Soluble dietary fiber is the portion of dietary fiber that can not be digested by digestive enzymes secreted by human digestive tract, but soluble in hot water, and its aqueous solution can be precipitated by four times ethanol. More and more scholars and people at home and abroad have paid more and more attention to the fermentation of seabuckthorn meal by Trichoderma leucovskii in order to improve the extraction rate of water-soluble dietary fiber of Hippophae rhamnoides and decolorize the water-soluble dietary fiber extracted from Hippophae rhamnoides after fermentation. The characteristics of water-soluble dietary fiber obtained from seabuckthorn were determined and analyzed. Through the screening of three fermentation strains, Trichoderma Richalii strain was found to be suitable for the solid fermentation of seabuckthorn meal. The factors affecting the preparation of water-soluble dietary fiber from Hippophae rhamnoides were inoculated at 5 mL 路100 mg / L and fermentation temperature at 28 鈩,
