【摘要】：建立一個(gè)復(fù)合擴(kuò)增體系,同時(shí)擴(kuò)增DYS391、GATA-A4、GATA-A10和GATA-H44個(gè)Y染色體特異性的STR基因座,并用聚丙烯酰胺凝膠電泳和銀染顯色技術(shù)進(jìn)行基因分型。采用該復(fù)合擴(kuò)增系統(tǒng)調(diào)查廣東漢族311名無(wú)關(guān)男性個(gè)體的單倍型頻率,這4個(gè)基因座分別檢出5、7、6和5個(gè)等位基因,其基因多樣性分別為0.4623、0.6972、0.7173、0.6015。共檢出98種單倍型,單倍型基因多樣性為0.9755。該復(fù)合擴(kuò)增體系在建立Y染色體STR數(shù)據(jù)庫(kù),研究群體遺傳和進(jìn)行法醫(yī)學(xué)鑒定有重要意義。
[Abstract]:A complex amplification system was established to amplify DYS391,GATA-A4,GATA-A10 and GATA-H44 Y chromosome specific STR loci at the same time. Polyacrylamide gel electrophoresis (page) and silver staining were used for genotyping. The haplotype frequencies of 311 unrelated male individuals in Guangdong Han population were investigated by the multiplex amplification system. The four loci were 5, 7, 6 and 5 alleles, respectively. The genetic diversity was 0.4623,0.6972, 0.7173 and 0.6015, respectively, and the genetic diversity was 0.4623,0.6972, 0.7173 and 0.6015, respectively. A total of 98 haplotypes were detected, and the gene diversity of haplotypes was 0.9755. The complex amplification system is of great significance in establishing Y chromosome STR database, studying population heredity and forensic identification.
