【摘要】：【目的】 應(yīng)用PCR擴(kuò)增、PAGE及銀染分型技術(shù)對(duì)常染色體STR基因座D5S818，D8S1179，D18S51及D21S11的遺傳學(xué)多態(tài)性進(jìn)行研究，獲取其中國(guó)吉林延邊地區(qū)朝鮮族群體遺傳學(xué)資料，并評(píng)估其法醫(yī)學(xué)應(yīng)用價(jià)值。 【方法】 應(yīng)用聚合酶鏈反應(yīng)(PCR)，聚丙烯酰胺凝膠電泳(PAGE)及銀染分型技術(shù)，對(duì)延邊地區(qū)109名朝鮮族無關(guān)個(gè)體進(jìn)行群體調(diào)查，獲得四種基因座的群體遺傳學(xué)資料。 【結(jié)果】 通過群體調(diào)查，在D5S818，D8S1179，D18S51及D21S11基因座中分別檢出8、8、14及12個(gè)等位基因和21、17、42及32種基因型。評(píng)估法醫(yī)學(xué)統(tǒng)計(jì)結(jié)果，雜合度觀察值(Ho)分別為0.835、0.780、0.884及0.716，期望值(He)分別為0.798、0.837、0.862及0.767；多態(tài)性信息量(PIC)分別為0.768，0.815，，0.848及0.739；個(gè)體識(shí)別能力(PD)分別為0.916、0.949、0.957及0.910；累積PD為99.9983％，累積匹配概率為1.658×10~(-5)；Hardy-Weinberg平衡檢驗(yàn)p值分別為0.421、0.640、0.811及0.837，均符合Hardy-Weinberg平衡。此外將延邊地區(qū)朝鮮族人群的等位基因分布頻率與其他不同種族、民族人群等位基因分布頻率之間進(jìn)行了比較。結(jié)果不同種族、民族之間存在著不同程度的差異。最終闡述了建立延邊地區(qū)朝鮮族人群STR基因座數(shù)據(jù)庫的必要性。 【結(jié)論】 本文所述四個(gè)基因座均屬于高鑒別力、高雜合度、高信息量的STR基因座，是在法醫(yī)學(xué)個(gè)人識(shí)別和親權(quán)鑒定中有較高應(yīng)用價(jià)值的遺傳標(biāo)記系統(tǒng)。
[Abstract]:[objective] to study the genetic polymorphisms of autosomal STR loci D5S818, D8S1179, D18S51 and D21S11 by PCR amplification, PAGE and silver staining. The genetic data of Korean population in Yanbian area, Jilin Province, China were obtained, and the value of forensic medicine application was evaluated. [methods] Polymerase chain reaction (PCR),) polyacrylamide gel electrophoresis (PAGE) and silver staining technique were used to investigate the population of 109 unrelated Korean individuals in Yanbian area. Population genetic data of four loci were obtained. [results] there were 8, 8, 14 and 12 alleles and 21, 17, 42 and 32 genotypes in D5S818, D8S1179, D18S51 and D21S11 loci, respectively. The observed values of heterozygosity (Ho) were 0.835,0.780,0.884 and 0.716, and the expected values of (He) were 0.798,0.837, 0.862 and 0.767, respectively. The polymorphic information content (PIC) was 0.768,0.815,0.848 and 0.739 respectively, the individual identification ability (PD) was 0.916,0.949, 0.957 and 0.910, the cumulative PD was 99.9983%, and the cumulative matching probability was 1.658 脳 10 ~ (- 5). The p values of Hardy-Weinberg equilibrium test were 0.421, 0.640, 0.811 and 0.837, respectively, which were in agreement with the Hardy-Weinberg equilibrium. In addition, the allelic frequency of Korean population in Yanbian area was compared with that of other ethnic groups and ethnic groups. Results there are different degrees of differences among different races and nationalities. Finally, the necessity of establishing the database of STR loci in Korean population in Yanbian area was expounded. [conclusion] the four loci described in this paper all belong to STR loci with high discriminative ability, high heterozygosity and high information content. It is a genetic marker system with high application value in forensic personal identification and paternity identification.
