法醫(yī)昆蟲種屬鑒定的分子生物學(xué)方法研究

【摘要】:目的 本課題旨在探索新的法醫(yī)昆蟲學(xué)種屬鑒定方法。方法 利用引物設(shè)計軟件分別對法醫(yī)常見昆蟲的常染色體微衛(wèi)星區(qū)域和線粒體DNA(mtDNA)進(jìn)行引物設(shè)計,使用PCR技術(shù)分別對其進(jìn)行擴(kuò)增,用聚丙烯酰胺電泳對PCR產(chǎn)物分離、銀染。對于微衛(wèi)星(microsatellite)遺傳標(biāo)記直接進(jìn)行分型檢測。對于線粒體DNA通過銀染分離、檢測,利用DNA測序儀進(jìn)行測序分析。用系統(tǒng)進(jìn)化軟件(Mega2.1和PHYLIP3.06)進(jìn)行法醫(yī)常見昆蟲的親緣關(guān)系分析。結(jié)果 采用對線粒體16rsRNA基因進(jìn)行PCR擴(kuò)增和DNA測序的方法,我們得到了中國9個地區(qū)6種不同法醫(yī)常見昆
[Abstract]:Objective to explore a new method for identification of forensic entomology species. Methods Primer design software was used to design primers for autosomal microsatellite region and mitochondrial DNA (mtDNA) of common insects in forensic science, PCR technique was used to amplify them, polyacrylamide electrophoresis was used to isolate PCR products and silver staining was used. The genotyping of microsatellite (microsatellite) markers was carried out directly. Mitochondrial DNA was separated and detected by silver staining and sequenced by DNA sequencer. Phylogenetic software (Mega2.1 and PHYLIP3.06) was used to analyze the phylogenetic relationship of common insects in forensic medicine. Results by means of PCR amplification and DNA sequencing of mitochondrial 16rsRNA gene, we obtained 6 kinds of common forensic medicine in 9 regions of China.
