彝族群體Y-STR和Y-SNP多態(tài)性的研究和法醫(yī)學(xué)應(yīng)用

【摘要】: 目的:Y染色體除擬常染色體區(qū)外,在遺傳過(guò)程中不發(fā)生染色體重組,呈父系單倍型遺傳特點(diǎn),因此應(yīng)用Y染色體多態(tài)性DNA遺傳標(biāo)記,可對(duì)不同群體間的遺傳距離進(jìn)行分析,從而揭示不同群體的起源、進(jìn)化、遷移和親緣關(guān)系等。同時(shí)在法醫(yī)學(xué)實(shí)踐中,Y染色體父系單倍型遺傳特點(diǎn),使其成為混合斑分析和父系親屬間親緣關(guān)系鑒定的重要工具。本研究還為中國(guó)彝族的基因信息提供了基礎(chǔ)數(shù)據(jù)。 方法:本研究從AmpFlSTR? Y filerTM系統(tǒng)中選擇了DYS456、DYS389Ⅰ、DYS390、DYS389Ⅱ、DYS458、DYS19、DYS385a,b、DYS393、DYS391、DYS439、DYS635、DYS392、Y GATA H4、DYS437、DYS438、DYS448共計(jì)17個(gè)Y-STR基因座,應(yīng)用PCR擴(kuò)增和毛細(xì)管電泳分離,得到Y(jié)-STR基因座分型。并從前期研究的課題“南方漢族群體Y-STR和Y-SNP系統(tǒng)的研究和法醫(yī)學(xué)應(yīng)用”中選擇了M111、M117、M119、M122、M134、M159、M168、M175、M214、M7、M88、M89、M9、M95、RPS4Y共15個(gè)頻率大于5%的SNP片段,應(yīng)用PCR擴(kuò)增直接測(cè)序方法得到16個(gè)SNP位點(diǎn)。研究了我國(guó)彝族64個(gè)樣本的Y-STR和Y-SNP擴(kuò)展單倍型。并應(yīng)用統(tǒng)計(jì)學(xué)方法與漢族(南漢,北漢)、回族、維族、蒙古族、藏族樣本的結(jié)果進(jìn)行比較。 結(jié)果:得到了彝族Y-STR和Y-SNP的等位基因及單倍型頻率多態(tài)性分布資料。64個(gè)男性無(wú)關(guān)個(gè)體,共檢測(cè)到了63種單倍型。17個(gè)Y-STR基因座共檢測(cè)到63種單倍型及90個(gè)等位基因,基因頻率分布在0.0159-0.8889之間,基因多樣性值(GD)分布在0.2007-0.9002之間;16個(gè)SNP位點(diǎn)共檢測(cè)到11種單倍型。 結(jié)論:經(jīng)典的直接測(cè)序法可準(zhǔn)確的檢測(cè)單個(gè)堿基的變異,可用于法醫(yī)學(xué)混合斑中男性成分的個(gè)體識(shí)別及父系親緣關(guān)系的鑒定。統(tǒng)計(jì)數(shù)據(jù)結(jié)果表明,彝族與回族、維族、蒙古族、北方漢族在Y染色體遺傳基因上的遺傳距離較近,而與藏族、南方漢族遺傳距離相對(duì)較遠(yuǎn)。
