【摘要】：目的：探討大鼠死后腦組織中兩種不同看家基因(18s rRNA和GAPDH mRNA)RNA含量比值(GAPDH mRNA／18s rRNA)變化與死后間隔時間的關(guān)系，并檢測是否具有統(tǒng)計學(xué)意義。旨為法醫(yī)實踐中推斷死亡時間提供科學(xué)的參考依據(jù)。 方法：采用CO_2窒息法處死健康成年SD大鼠建立動物模型，并將其分別放置于恒定環(huán)境濕度為30％，但不同溫度(15℃和20℃)環(huán)境的兩組實驗分組。采用TRIzol試劑提取SD大鼠在死后0d、1d、2d、3d、5d、7d時腦組織中的總RNA。采用RT-PCR方法與凝膠圖像分析技術(shù)檢測18s rRNA和GAPDH mRNA的含量，并將二者的比值(GAPDH mRNA／18s rRNA)與死后間隔時間進行統(tǒng)計學(xué)回歸分析。 結(jié)果：(1)在兩組實驗分組中，直到2d時死后大鼠腦組織中GAPDH mRNA降解不明顯；但在3d、5d、7d時腦組織中GAPDH mRNA降解程度顯著增加。而18S rRNA直到7d仍無顯著性降解變化。(2)大鼠死后腦組織中看家基因GAPDH mRNA降解受到溫度的影響——隨著溫度的升高，降解速度增加。而18S rRNA降解緩慢，幾乎不受到溫度的影響。(3)對大鼠死后腦組織內(nèi)GAPDH mRNA／18s rRNA值(Y)變化與死后間隔時間(X)進行統(tǒng)計學(xué)回歸分析：在20℃組中，Y=0.62-0.004X(R=-0.981，，P＜0.001)；在15℃組中，Y=0.965-0.001X(R=-0.709，P＜0.001)。GAPDH mRNA／18s rRNA值與死后間隔時間之間存在顯著相關(guān)性。 結(jié)論：在本實驗中，隨著死后間隔時間的延長SD大鼠腦組織中GAPDH mRNA發(fā)生降解。其20℃時降解速度要快于15℃，而18S rRNA降解緩慢，幾乎不受到溫度的影響。GAPDH mRNA／18s rRNA與死后間隔時間存在明顯相關(guān)性，可望作為推斷死亡時間的參考指標之一。
