廣西都安瑤族線粒體DNA控制區(qū)及Y-STR多態(tài)性研究

【摘要】:第一部分 廣西都安瑤族線粒體DNA控制區(qū)多態(tài)性研究 目的 為線粒體DNA(mitochondrial DNA, mtDNA)法醫(yī)學(xué)應(yīng)用提供群體遺傳學(xué)數(shù)據(jù),擴大mtDNA在個體識別中的應(yīng)用范圍。方法 以南方民族瑤族為研究對象,根據(jù)mtDNA控制區(qū)及周圍區(qū)域的序列設(shè)計引物,用PCR產(chǎn)物直接測序?qū)?05份無關(guān)瑤族樣本mtDNA控制區(qū)進行了測序,通過Sequencing Analysis 3.4和Seq/Ede軟件分析比對,得到的序列結(jié)果,計算瑤族mtDNA單倍型的識別率,并應(yīng)用MEGA 2軟件對數(shù)據(jù)做進化距離分析,構(gòu)建系統(tǒng)發(fā)育樹。結(jié)果 對樣本的mtDNA控制區(qū)序列進行了統(tǒng)計分析,共檢出141個變異點,平均變異數(shù)10.5個,發(fā)現(xiàn)3處點異質(zhì)性。整個序列個體識別率為0.9747,變異度為0.9841。除了HVⅠ、HVⅡ及HVⅢ外,發(fā)現(xiàn)瑤族mtDNA控制區(qū)還存在其它的相對高變區(qū),幾錷tDNA控制區(qū)序列與其他民族相比,均存在一定差異。系統(tǒng)發(fā)育樹顯示,瑤族大體出現(xiàn)5個聚類簇,每個聚類簇各有特點。結(jié)論 瑤族mtDNA控制區(qū)檢出3個點異質(zhì)性,并發(fā)現(xiàn)存在其他相對高變區(qū)。群體遺傳學(xué)數(shù)據(jù)提示瑤族與蒙古人種有較近的親緣關(guān)系。本研究為法醫(yī)學(xué)及系統(tǒng)發(fā)育學(xué)研究提供了基礎(chǔ)的群體遺傳學(xué)數(shù)據(jù)。
[Abstract]:The first part of the study on mitochondrial DNA control region polymorphism of the Yao nationality in Duan Guangxi aims to provide population genetic data for the forensic application of mitochondrial DNA (mitochondrial DNA, mtDNA).
