三個(gè)STR基因座熒光標(biāo)記復(fù)合擴(kuò)增的研究及其法醫(yī)學(xué)應(yīng)用

【摘要】:目的 本課題旨在尋找新的有應(yīng)用價(jià)值的STR基因座,并建立D12S1064、GATA140E03、D18S536基因座的熒光標(biāo)記復(fù)合擴(kuò)增體系。按照美國(guó)DNA分析方法技術(shù)工作組(TWGDAM)的指導(dǎo)方案,進(jìn)行了法醫(yī)學(xué)應(yīng)用性研究。方法 首先對(duì)5個(gè)新的STR基因座,即D9S1119、D12S1064、D18S536、GATA156D11和GATA140E03,進(jìn)行群體遺傳學(xué)研究。從中獲得三個(gè)多態(tài)性好、個(gè)人識(shí)別機(jī)率高的三個(gè)基因座,,即D12S1064、GATA140E03、D18S536,利用傳統(tǒng)的復(fù)合擴(kuò)增的方法,用美國(guó)ABI-310毛細(xì)管電泳儀進(jìn)行熒光標(biāo)記復(fù)合擴(kuò)增的研究。并對(duì)復(fù)合擴(kuò)增系統(tǒng)的靈敏度和準(zhǔn)確度,基因座的種屬特異性,不同退火溫度下復(fù)合擴(kuò)增的效果等進(jìn)行了重復(fù)性研究。結(jié)果 獲得了5個(gè)STR基因座的基因頻率、個(gè)人識(shí)別率、非父排除率等法醫(yī)鑒定中所需要的遺傳學(xué)數(shù)據(jù);成功建立了D12S1064、GATA140E03、
[Abstract]:Objective to search for a new STR locus and to establish a fluorescent multiplex amplification system for D12S1064-GATA140E0E03 D18S536 locus. The application of forensic science was studied according to the guidance of (TWGDAM) of the DNA Analytical Methodology Technical working Group in the United States. Methods five new STR loci, D9S1119, D12S1064, D18S536, GATA156D11 and GATA140E03, were studied in population genetics. Three loci, D12S1064, GATA140E03, D18S536, which have good polymorphism and high probability of personal identification, were obtained. The fluorescence labeling multiplex amplification was carried out by ABI-310 capillary electrophoresis in the United States. The sensitivity and accuracy of the multiplex amplification system, the specificity of gene loci and the effect of multiple amplification at different annealing temperatures were studied repeatedly. Results genetic data of 5 STR loci such as gene frequency, personal identification rate, non-paternal exclusion rate and so on, were obtained, and D12S1064-GATA140E03 was successfully established.
