【摘要】:目的探討熒光半定量逆轉(zhuǎn)錄-聚合酶鏈?zhǔn)椒磻?yīng)(fluorescencesemi-quantitativereversetranscrip-tase-polymerasechainreaction,RT-PCR)技術(shù)檢測大鼠死后不同時(shí)間腎3-磷酸甘油醛脫氫酶(GAPDH)mRNA含量變化對死亡時(shí)間(postmorteminterval,PMI)推斷的應(yīng)用價(jià)值。方法28只大鼠斷頸處死后置于20℃溫控系統(tǒng)內(nèi),利用熒光標(biāo)記半定量RT-PCR技術(shù)檢測大鼠腎GAPDHmRNA在死后48h內(nèi)相對表達(dá)量的變化情況。結(jié)果在死后即刻至40h內(nèi)大鼠腎均可檢測出GAPDHmRNA,且其擴(kuò)增產(chǎn)物呈規(guī)律性下降。結(jié)論熒光半定量RT-PCR技術(shù)檢測大鼠死后腎GAPDHmRNA含量變化對PMI推斷具有一定的應(yīng)用價(jià)值。
