【摘要】:前言 Rh血型系統(tǒng)是最為復(fù)雜、最具多態(tài)性的人類血型系統(tǒng)之一,具有重要的臨床意義。同時(shí),作為一種優(yōu)良的遺傳標(biāo)記,Rh血型從發(fā)現(xiàn)的時(shí)候開(kāi)始就被廣泛應(yīng)用于法醫(yī)學(xué)實(shí)踐。目前中國(guó)的Rh血型分型方法是傳統(tǒng)的血清學(xué)技術(shù),這種技術(shù)在實(shí)際應(yīng)用中存在著種種弊端。比如:實(shí)驗(yàn)多依賴價(jià)格昂貴的進(jìn)口抗血清,實(shí)驗(yàn)結(jié)果的判斷易受多種因素的影響,難以鑒別稀有的Rh變異型等。特別對(duì)于產(chǎn)前親子鑒定和曾經(jīng)接受過(guò)多次輸血人員的血型鑒定與親子鑒定等特殊的法醫(yī)學(xué)問(wèn)題,傳統(tǒng)方法更是束手無(wú)策。近年來(lái),隨著Rh系統(tǒng)分子生物學(xué)研究的深入,RH基因分型技術(shù)應(yīng)運(yùn)而生,為解決以上難題提供了新的思路,成為研究熱點(diǎn)。目前國(guó)內(nèi)尚無(wú)應(yīng)用基因分型技術(shù)檢測(cè)Rh血型的相關(guān)報(bào)道,而RH基因在不同人種間的變異使國(guó)外的研究成果不能直接應(yīng)用于中國(guó)人群。因此,本研究應(yīng)用序列特異性引物PCR(PCR-SSP)技術(shù)對(duì)兩個(gè)不同的中國(guó)群體進(jìn)行RHCE基因型分布的調(diào)查,旨在探討這一新興技術(shù)的應(yīng)用價(jià)值。 材料和方法 1.樣品來(lái)源 1.1 群體調(diào)查樣品 200例中國(guó)北方漢族健康、無(wú)關(guān)個(gè)體血樣由中國(guó)醫(yī)科大學(xué)法醫(yī)學(xué)院提供,200例中國(guó)南方黎族健康、無(wú)關(guān)個(gè)體血樣由海南醫(yī)學(xué)院提供。 1.2 親子鑒定樣品 5例親子鑒定案件樣品由中國(guó)醫(yī)科大學(xué)法醫(yī)學(xué)院提供。 2.實(shí)驗(yàn)方法 2.1 Rh血型血清學(xué)分型:應(yīng)用單克隆抗-D、抗-C、抗-c、抗-E及
[Abstract]:Preface Rh blood group system is one of the most complex and polymorphic human blood group systems, which has important clinical significance. At the same time, as an excellent genetic marker, Rh blood group has been widely used in forensic practice since its discovery. At present, the Rh blood group typing method in China is a traditional serological technique, which has many disadvantages in practical application. For example, the experiment relies on the expensive imported antiserum, and the judgement of the experimental results is easily influenced by many factors, so it is difficult to identify the rare Rh variant and so on. Especially for the special forensic problems such as prenatal paternity testing and blood type identification and paternity testing of people who have received multiple blood transfusions, the traditional methods are even more helpless. In recent years, with the in-depth study of Rh molecular biology, RH genotyping technology emerged as the times require, which provides a new way to solve the above problems and becomes a research hotspot. At present, there are no reports on the application of genotyping technique in the detection of Rh blood group in China, but the variation of RH gene in different ethnic groups makes it impossible for foreign research results to be directly applied in Chinese population. Therefore, the RHCE genotypes of two different Chinese populations were investigated by sequence specific primer PCR-SSPP, in order to explore the application value of this new technique. Materials and methods 1. Sample source 1.1 Group Survey samples The unrelated individual blood samples were provided by the School of Forensic Medicine of China Medical University in 200 cases of Li nationality health in southern China, and the unrelated individual blood samples were provided by Hainan Medical College. 1.2 paternity test samples The samples of 5 cases of paternity test were provided by the School of Forensic Medicine of China Medical University. 2.
