弓形蟲感染對小鼠腦內(nèi)c-fos及ACTH表達(dá)的影響

【摘要】: 本文通過尾靜脈注射方式使KM小鼠感染弓形蟲,觀察大腦皮質(zhì)、腦干、間腦、中腦及海馬組織中弓形蟲的動(dòng)態(tài)分布,同時(shí),應(yīng)用免疫組織化學(xué)技術(shù)觀察原癌基因c-fos及ACTH在腦組織中的表達(dá)。探討在神經(jīng)內(nèi)分泌免疫網(wǎng)絡(luò)中:腎上腺皮質(zhì)激素(ACTH)和c-fos在弓形蟲病腦組織中的表達(dá)及其意義。 實(shí)驗(yàn)結(jié)果表明:小鼠尾靜脈感染弓形蟲的第8天,大腦皮質(zhì)和間腦中觀察到弓形蟲滋養(yǎng)體;第10天,在腦干、中腦及海馬中查獲蟲體。②免疫組織化學(xué)染色檢測顯示:腦組織內(nèi)ACTH表達(dá)明顯,呈陽性反應(yīng)的ACTH胞體出現(xiàn)在大腦皮質(zhì)及下丘腦區(qū)內(nèi),ACTH免疫反應(yīng)陽性纖維出現(xiàn)在下丘腦及胼胝體內(nèi),大量而密集的ACTH免疫反應(yīng)陽性纖維主要集中在下丘腦第三腦室室周區(qū)、室旁核、弓狀核及其相鄰?fù)鈧?cè)區(qū)域內(nèi)。③在弓形蟲感染小鼠的大腦腦組織內(nèi)有c-fos的表達(dá),但與對照組比較,出現(xiàn)明顯差異,大腦皮質(zhì)神經(jīng)元胞體內(nèi)出現(xiàn)c-fos免疫反應(yīng)陽性顆粒,染色強(qiáng)度明顯低于對照組。海馬的神經(jīng)元胞體內(nèi)未見有c-fos免疫反應(yīng)陽性顆粒。下丘腦神經(jīng)元胞體內(nèi)則出現(xiàn)含量較多的免疫反應(yīng)陽性顆粒,且顯色強(qiáng)度高于對照組,這些免疫反應(yīng)陽性顆粒主要集中于大神經(jīng)細(xì)胞內(nèi)。 結(jié)論:①弓形蟲(RH株)靜脈途徑感染小鼠,蟲體通過血腦屏障侵入腦組織;腦組織區(qū)域不同,血腦屏障對弓形蟲的阻隔作用存在差異。②感染弓形蟲小鼠腦中,大腦皮質(zhì)和下丘腦的神經(jīng)元中ACTH的表達(dá)明顯增強(qiáng)。③感染弓形蟲小鼠,在海馬和大腦皮質(zhì)c-fos蛋白表達(dá)較正常組顯著降低,而在下丘腦區(qū)c-fos蛋白表達(dá)則明顯高于正常對照組。
[Abstract]:In this paper, km mice were infected with Toxoplasma gondii by tail vein injection, and the dynamic distribution of Toxoplasma gondii in cerebral cortex, brainstem, diencephalon, midbrain and hippocampus was observed. The expression of proto-oncogene c-fos and ACTH in brain tissue was observed by immunohistochemical technique. To investigate the expression and significance of ACTH and c-fos in neuroendocrine and immune networks in Toxoplasma gondii. The results showed that Toxoplasma gondii trophozoites were observed in the cerebral cortex and diencephalon on the 8th day of Toxoplasma gondii infection in the tail vein of mice, and in the brain stem on the 10th day. Immunohistochemical staining showed that the expression of ACTH was obvious in the brain and hippocampus. The positive ACTH cells appeared in the hypothalamus and corpus callosum, and a large number of ACTH immunoreactive fibers were concentrated in the periventricular area of the hypothalamus. The expression of c-fos in paraventricular nucleus, arcuate nucleus and adjacent lateral region was found in the brain tissue of mice infected with Toxoplasma gondii, but there was significant difference between the control group and the control group. C-fos immunoreactive granules appeared in the cortical neurons. The staining intensity was obviously lower than that of the control group. No c-fos immunoreactive granules were found in the neuronal bodies of hippocampus. There were more immunoreactive granules in hypothalamic neurons, and the color intensity was higher than that in the control group. These immunoreactive granules were mainly concentrated in large nerve cells. Conclusion the mice infected with Toxoplasma gondii (RH strain) via venous pathway, the parasites invade into the brain tissue through blood-brain barrier, and the barrier effect of blood-brain barrier to Toxoplasma gondii is different in mice with different brain tissue regions. 2. Infection of Toxoplasma gondii in mice with Toxoplasma gondii. The expression of ACTH in neurons of cerebral cortex and hypothalamus was significantly increased in mice infected with Toxoplasma gondii. The expression of c-fos protein in hippocampus and cerebral cortex was significantly lower than that in normal group, but the expression of c-fos protein in hypothalamus was significantly higher than that in normal control group.
