三重腦震蕩鼠腦色氨酸羥化酶及酪氨酸羥化酶變化研究

【摘要】: 目的:建立三重腦震蕩(multiple cerebral concussion,MCC)大鼠模型,觀察MCC大鼠腦干色氨酸羥化酶(Tryptophan hydroxylase,TPH)與基底節(jié)和中隔區(qū)酪氨酸羥化酶(tyrosine hydroxylase,TH)表達(dá)變化情況,探討與MCC大鼠認(rèn)知行為障礙相關(guān)的色氨酸羥化酶和酪氨酸羥化酶變化的關(guān)系,以了解三重腦震蕩鼠認(rèn)知行為障礙的神經(jīng)生物學(xué)基礎(chǔ)。方法:采用自制金屬單擺式閉合性腦損傷機(jī)械打擊裝置成功復(fù)制腦震蕩大鼠模型48只;大鼠隨機(jī)分為1天、2天、4天、8天、16天和24天6個三重腦震蕩組(n=8)和1個正常對照組(Control,CON,n=8)。應(yīng)用免疫印跡技術(shù)(Western Blotting)分別檢測大鼠腦震蕩損傷后第1、2、4、8、16、24天腦干色氨酸羥化酶(Tryptophanhydroxylase,TPH)以及基底節(jié)和中隔區(qū)酪氨酸羥化酶(Tyrosine hydroxylase,TH)的表達(dá)變化規(guī)律。結(jié)果:免疫印跡結(jié)果顯示,腦干TPH表達(dá)自傷后1天起,呈逐漸上調(diào)趨勢,到傷后第2天該酶的表達(dá)到最高峰,傷后2-8天的表達(dá)升高,與對照組相比較有顯著性差異(p<0.05)。傷后16天,TPH表達(dá)逐漸下降,到傷后第24天基本已經(jīng)回落到對照組的水平。基底節(jié)TH在傷后表達(dá)總體呈上升趨勢,到傷后第4天該酶的表達(dá)到最高峰,與對照組相比較,傷后2-16天均有顯著性差異(p<0.05)。中隔區(qū)TH在傷后表達(dá)總體呈上升趨勢,傷后第1天開始上升,到傷后第4天該酶的表達(dá)到最高峰,8天和16天略有下降,24天再次升高,傷后第4天和第24天的表達(dá)與對照組相比較有顯著性差異(p<0.05)。結(jié)論:1、三重腦震蕩大鼠傷后腦干色氨酸羥化酶的表達(dá)升高。2、三重腦震蕩大鼠傷后基底節(jié)、中隔區(qū)酪氨酸羥化酶的表達(dá)升高。3、根據(jù)本課題組之前的研究以及本次研究結(jié)果推測三重腦震蕩后大鼠的認(rèn)知缺陷可能與腦干TPH及基底節(jié)和中隔區(qū)TH的表達(dá)升高有關(guān)。
[Abstract]:Objective: to establish a rat model of multiple cerebral concussionMCCs with triple cerebral concussion, and to observe the expression of Tryptophan hydroxylase (Tryptophan hydroxylase) and tyrosine hydroxylase tyrosine hydroxylase (THH) in the basal ganglia and septum of MCC rats. To explore the relationship between the changes of tryptophan hydroxylase and tyrosine hydroxylase associated with cognitive behavioral disorders in MCC rats and to understand the neurobiological basis of cognitive behavioral disorders in rats with triple concussion. Methods: Forty-eight rats with cerebral concussion were randomly divided into 6 concussion group (n = 6) and a control group (n = 48) and a normal control group (n = 48), which were randomly divided into two groups: 1 day, 2 days, 4 days, 8 days, 16 days, and 24 days, 6 triple concussion groups. Western blotting technique was used to detect the expression of tryptophan hydroxylase (Tryptophan hydroxylase), tyrosine hydroxylase (Tyrosine hydroxylaseTHH) and tyrosine hydroxylase (Tyrosine hydroxylaseTH3) in brain stem of rats after cerebral concussion injury. Results: the results of Western blotting showed that the expression of TPH in the brain stem increased gradually from 1 day after injury to the peak on the second day after injury, and increased at 2-8 days after injury. There was a significant difference between the two groups in comparison with the control group (P < 0.05). The expression of TPH decreased gradually on the 16th day after injury, and decreased to the level of the control group on the 24th day after injury. The expression of th in basal ganglia showed a general upward trend after injury, and the expression of th reached its peak on the 4th day after injury. Compared with the control group, the expression of th in basal ganglia was significantly different from that of the control group on the 2-16 days after injury (P < 0.05). The expression of th in septal area showed a general upward trend after injury, which began to rise on the first day after injury, and increased again on the 4th day after injury to the peak at the peak of 8 days and 16 days after injury, and then increased again at 24 days after injury. On the 4th and 24th day after injury, there was significant difference between the control group and the control group (p < 0.05). Conclusion the expression of tryptophan hydroxylase in brain stem of rats with brain stem concussion was increased. The expression of tyrosine hydroxylase in septum was increased. According to our previous study and the results of this study, we speculated that the cognitive deficiency of rats after triple concussion might be related to the increased expression of TPH and th in basal ganglia and septum.
