【摘要】： 背景EB病毒是被明確列為致癌因子的一種γ皰疹病毒，它與鼻咽癌、B細胞淋巴瘤、霍奇金氏病、伯基特淋巴瘤等多種惡性腫瘤密切相關。Rta蛋白是EB病毒裂解期立即早期基因之一BRLF1的編碼產(chǎn)物，是EB病毒由潛伏期轉向裂解復制的關鍵調(diào)控因子。它能引起一系列裂解早期基因的“瀑布式表達”，最終引發(fā)EB病毒裂解傳染。目前Rta蛋白已成為國外研究的重點。 目的以特異性的抗Rta多克隆抗體體作為探針，初步研究Rta在胃癌(gastric carcinoma)、食管癌(esophageal carcinoma)、乳腺癌(breast carcinoma)和肺癌(lung carcinoma)中的表達情況。 方法采用免疫組織化學的方法(Envision二步法)，將純化好的多克隆抗體作為一抗，檢測了45例食管癌、30例胃癌、42例乳腺癌和32例肺癌組織樣本中Rta蛋白的表達情況。 結果45例食管癌中顯示對抗Rta抗體反應陽性者6例，相應癌旁組織均為陰性，P=0.026；30例胃癌中顯示對抗Rta抗體反應陽性者6例，相應癌旁組織均為陰性，P=0.031；42例乳腺癌中顯示對抗Rta抗體反應陽性者7例，相應癌旁組織均為陰性，，P=0.012；32例肺癌中顯示對抗Rta抗體反應陽性者4例，相應癌旁組織均為陰性，P=0.113。 結論EB病毒的裂解感染狀態(tài)與食管癌、胃癌和乳腺癌的發(fā)生具有一定相關性，裂解感染期EBV可能在上述惡性腫瘤的發(fā)病中起了一定作用。
[Abstract]:The background of EB virus is specified as a gamma herpesvirus carcinogen, and nasopharyngeal carcinoma, B cell lymphoma, Hodge's disease, Burkitt's lymphoma and other malignant tumors is closely related to.Rta protein of EB virus is cracking immediately one of the early gene BRLF1 encoding product, EB is a key regulator of virus from latency to lytic replication. It can cause a series of early cracking "waterfall gene expression", eventually lead to cracking of EB virus infection. The Rta protein has become the focus of research.
Objective to investigate the expression of Rta in gastric cancer (gastric carcinoma), esophageal cancer (esophageal carcinoma), breast cancer (breast carcinoma) and lung cancer (lung carcinoma) by using the specific anti Rta polyclonal antibody as a probe.
Methods using immunohistochemical method (Envision two step method), the purified polyclonal antibody was used as an antibody to detect the expression of Rta protein in 45 cases of esophageal cancer, 30 cases of gastric cancer, 42 cases of breast cancer and 32 cases of lung cancer.
Results 45 cases of esophageal carcinoma showed anti Rta antibody positive in 6 cases, paracancerous tissues were negative for P=0.026; in 30 cases of gastric carcinoma showed anti Rta antibody positive in 6 cases, paracancerous tissues were negative for P=0.031; in 42 cases of breast cancer showed anti Rta antibody positive in 7 cases and the corresponding paracancerous tissues were negative for P=0.012; 32 cases of lung cancer showed anti Rta antibody positive in 4 cases, paracancerous tissues were negative for P=0.113.
Conclusion there is a certain correlation between the infection status of EB virus and the occurrence of esophageal cancer, gastric cancer and breast cancer. EBV may play a role in the pathogenesis of these malignant tumors.
