【摘要】： 本課題建立了固相微萃取、固相萃取以及傳統(tǒng)的液-液萃取結(jié)合氣相色譜-質(zhì)譜聯(lián)用法分析了多種新型濫用藥物(2C-B、尼美西泮、硝基安定、咪噠唑侖、曲馬多、美沙酮和苯丙胺類)在生物體內(nèi)的原體藥物及其代謝產(chǎn)物的定性、定量檢測(cè)方法。實(shí)驗(yàn)以Wistar大鼠和人為生物模型,口服上述藥物,收集24小時(shí)內(nèi)的尿液和提取生物組織(肝、腦、血液、毛發(fā)等)作為生物檢材,利用固相微萃取法從生物檢材中提取了曲馬多、美沙酮,考察了固相微萃取是的萃取溫度、萃取時(shí)間、pH以及離子強(qiáng)度等實(shí)驗(yàn)條件;固相萃取法提取了多種生物檢材中的2C-B、尼美西泮、硝基安定、咪噠唑侖及其它們代謝產(chǎn)物;利用液-液提取法直接快速提取了檢材中的曲馬多和苯丙胺類濫用藥物及其它們的代謝產(chǎn)物。結(jié)合氣相色譜-質(zhì)譜聯(lián)用法定性或定量分析了上述多種濫用藥物及其代謝產(chǎn)物,實(shí)驗(yàn)定量分析了尿液中的曲馬多、美沙酮和毛發(fā)中苯丙胺類濫用藥物,定性檢測(cè)了2C-B、尼美西泮、硝基安定、咪噠唑侖、曲馬多、美沙酮在生物體內(nèi)的代謝產(chǎn)物,根據(jù)實(shí)驗(yàn)結(jié)果推斷了相關(guān)濫用藥物在生物體內(nèi)的代謝途徑,建立了檢測(cè)方法和相關(guān)的數(shù)據(jù)庫。建立的實(shí)驗(yàn)方法已經(jīng)成功運(yùn)用于實(shí)際工作中,并取得了良好的效果。
[Abstract]:This paper established a solid phase microextraction, solid phase extraction and conventional liquid-liquid extraction combined with gas chromatography-mass spectrometry analysis of a variety of new drug abuse (2C-B, nimetazepam and nitrazepam, midazolam, tramadol, methadone and amphetamines) qualitative parent drug and its metabolites in vivo the quantitative detection method. Experiments in Wistar rat and human biological model, the oral drug, urine was collected within 24 hours and the extraction of biological tissues (liver, brain, blood, hair, etc.) as biological samples by SPME extracted from biological samples of tramadol, methadone, effects of extraction temperature, solid phase micro extraction extraction time, pH and ionic strength conditions; solid-phase extraction of a variety of biological samples in 2C-B, nimetazepam and nitrazepam, midazolam and their metabolites; using liquid-liquid extraction The method of direct rapid extraction samples in the tramadol and abuse of amphetamine type drugs and their metabolites. Combined with GC-MS qualitative or quantitative analysis of the various abuse of drugs and their metabolites, the quantitative analysis of urine in tramadol abuse of amphetamine type drugs, methadone and hair in the qualitative detection of 2C-B, nimetazepam and nitrazepam, midazolam, tramadol, metabolites of methadone in organism, according to the experimental results that the metabolic pathways related to drug abuse in the organism, established the detection method and the related database. The established method has been successfully applied to the practical work, and have achieved good results.
