體外負(fù)壓培養(yǎng)對(duì)小鼠骨髓間充質(zhì)干細(xì)胞增殖能力和血管內(nèi)皮生長(zhǎng)因子分泌水平的影響

背景:如何增強(qiáng)骨髓間充質(zhì)干細(xì)胞增殖活性使其移植后發(fā)揮應(yīng)有的療效是目前急需解決的問(wèn)題。目的:探討體外負(fù)壓培養(yǎng)技術(shù)對(duì)小鼠骨髓間充質(zhì)干細(xì)胞增殖活性及血管內(nèi)皮生長(zhǎng)因子分泌水平的影響。方法:取第3代骨髓間充質(zhì)干細(xì)胞,給予間歇性負(fù)壓培養(yǎng)(-6.65,-13.3,-26.6 kPa),2 h/次,1次/12 h,對(duì)照組在正常條件下培養(yǎng)。培養(yǎng)12,24,36,48,60 h,采用CCK-8法檢測(cè)細(xì)胞增殖情況,ELISA檢測(cè)細(xì)胞分泌血管內(nèi)皮生長(zhǎng)因子水平,RT-PCR檢測(cè)血管內(nèi)皮生長(zhǎng)因子受體mRNA表達(dá)。根據(jù)上述結(jié)果,選擇一個(gè)最佳負(fù)壓條件和時(shí)間(-26.6 kPa,24 h),將骨髓間充質(zhì)干細(xì)胞分為正常對(duì)照組、負(fù)壓組、負(fù)壓+血管內(nèi)皮生長(zhǎng)因子受體抑制劑Axitinib組,CCK-8法檢測(cè)細(xì)胞增殖情況,EDU試劑盒檢測(cè)EDU細(xì)胞陽(yáng)性率,結(jié)晶紫染色觀察細(xì)胞集落形成單位。結(jié)果與結(jié)論:①與對(duì)照組比較,3個(gè)負(fù)壓干預(yù)組骨髓間充質(zhì)干細(xì)胞顯著增殖(P<0.05);②與對(duì)照組比較,3個(gè)負(fù)壓干預(yù)組細(xì)胞分泌血管內(nèi)皮生長(zhǎng)因子水平顯著增高(P<0.05),血管內(nèi)皮生長(zhǎng)因子受體mRNA表達(dá)顯著增高(P<0.05);③...
