Ca 2+ /CaMKⅡ介導(dǎo)雄激素對(duì)HT22細(xì)胞突觸蛋白PSD95的快速影響

雄激素作為人體中一種重要的類固醇類激素,參與了許多發(fā)育和生理過(guò)程。雄激素的經(jīng)典作用途徑是通過(guò)與胞質(zhì)中的雄激素受體(Androgen Receptor,AR)結(jié)合,形成激素-受體復(fù)合物后,被轉(zhuǎn)運(yùn)至胞核,與DNA上的反應(yīng)元件相互作用,介導(dǎo)靶基因的轉(zhuǎn)錄調(diào)節(jié),該途徑作用緩慢,需要時(shí)間較長(zhǎng)。此外,研究證實(shí)雄激素還可快速激活胞內(nèi)的信號(hào)傳導(dǎo)分子,通過(guò)非AR依賴途徑發(fā)揮相關(guān)效應(yīng)。該作用的特點(diǎn)是速度較快,通常在數(shù)秒至數(shù)分鐘內(nèi)完成,而且不依賴于AR介導(dǎo)的轉(zhuǎn)錄-翻譯過(guò)程而直接產(chǎn)生相應(yīng)效應(yīng)。例如,雄激素可以快速使腎臟遠(yuǎn)端小管、免疫T細(xì)胞、人冠狀動(dòng)脈內(nèi)皮細(xì)胞內(nèi)Ca2+濃度增加等。這種現(xiàn)象被稱為雄激素的非基因組效應(yīng)。Ca2+作為普遍存在的第二信使之一,可廣泛調(diào)節(jié)細(xì)胞內(nèi)的效應(yīng),包括細(xì)胞增殖、細(xì)胞凋亡、細(xì)胞遷移及基因表達(dá)等。研究已證實(shí)Ca2+參與了長(zhǎng)時(shí)程增強(qiáng)(long-term potentiation,LTP)和長(zhǎng)時(shí)程抑制(long-term depression,LTD)現(xiàn)象。LTP和LTD被認(rèn)為是與學(xué)習(xí)記憶密切相關(guān)的神經(jīng)突觸可塑性的生物學(xué)基礎(chǔ)。本...
