醋酸鹽兼養(yǎng)培養(yǎng)下蛋白小球藻油脂積累的研究

【摘要】:以醋酸鹽作為蛋白小球藻兼養(yǎng)生長中的有機(jī)碳源,采用尼羅紅三維熒光-區(qū)域體積積分法測定中性脂含量,研究了蛋白小球藻生長過程中的中性脂積累規(guī)律,分析了兼養(yǎng)和自養(yǎng)條件下生物量產(chǎn)率、總脂產(chǎn)率以及油脂的脂肪酸組成差異.結(jié)果顯示,加入醋酸鈉后,蛋白小球藻即進(jìn)入對數(shù)增長期,生長迅速,且同時啟動中性脂的積累.5g/L醋酸鈉既能促使藻細(xì)胞生長和中性脂的持續(xù)積累,又能避免過高的有機(jī)碳源所帶來的環(huán)境脅迫,培養(yǎng)12d后,單位細(xì)胞中性脂含量達(dá)到自養(yǎng)條件下對照樣的4.5倍,生物量和總脂產(chǎn)率分別是對照樣的2.1倍和3.1倍.5g/L醋酸鈉作用下,油脂脂肪酸以飽和脂肪酸C16:0(22.42%)和不飽和脂肪酸C18:1(31.58%)、C18:2(10.89%)為主.
[Abstract]:Using acetate as the organic carbon source of Chlorella protein Chlorella, the neutral lipid accumulation in the growth process of Chlorella protein Chlorella was studied by means of the method of three dimensional fluorescence and regional volume integration of Nilored, which was used to determine the content of neutral lipid in the growth process of Chlorella vulgaris. The biological yield, total fat yield and fatty acid composition were analyzed. The results showed that when sodium acetate was added, Chlorella protein Chlorella entered the logarithmic growth period, grew rapidly and initiated the accumulation of neutral lipid at the same time. Sodium 5g/L acetate could not only promote the growth of algal cells and the accumulation of neutral lipid, but also promote the accumulation of neutral lipid. After being cultured for 12 days, the content of neutral lipid per unit cell was 4.5 times higher than that under autotrophic condition. The biomass and total fat yield were 2.1 times and 3.1 times higher than those of the control, respectively. Under the action of 5g/L sodium acetate, the fatty acids of oil and fat were C 16: 0 (22.42%) and C 18: 1 (31.58%), respectively. C18: 2 (10.89%).
