菌株P.aeruginosa BC1的篩

【摘要】:為改善克拉瑪依油田的微生物強化原油開采,從克拉瑪依油田廢水中篩選出一株性能優(yōu)良的生物表面活性劑生產菌株BC1。根據(jù)菌株BC1的理化指標和16S r DNA對其進行了屬種信息鑒定,通過薄層層析(TLC)、傅里葉紅外色譜(FT-IR)及高效液相色譜/質譜(HPLC-MS)對其產生物表面活性劑進行了分析,通過單因素搖瓶實驗考察了發(fā)酵條件對菌株BC1發(fā)酵合成生物表面活性劑的影響,研究了生物表面活性劑的性能。研究結果表明,菌株BC1為銅綠假單胞菌屬(Pseudomonas aeruginosa),其產生物表面活性劑主要成分為單鼠李糖脂Rh C10C10;在溫度37℃、初始p H為8、初始葡萄糖濃度5 g/L的條件下持續(xù)發(fā)酵96 h后,用酸沉降法測得鼠李糖脂產量為0.788 g/L;菌株BC1產生物表面活性劑能將發(fā)酵液表面張力從72降至28.6 m N/m,對車用機油的乳化指數(shù)E24高達93%。人工油砂剝油實驗中,發(fā)酵液、發(fā)酵離心上清液和鼠李糖脂粗品溶液對人工油砂的剝油率均達到74%以上。菌株P.aeruginosa BC1產生物表面活性劑在微生物強化采油等方面具有良好的應用潛能。
[Abstract]:In order to improve microbial enhanced crude oil production in Karamay Oilfield, a strain BC1 with excellent performance was screened from the wastewater of Karamay Oilfield. According to the physicochemical index of strain BC1 and the information of 16s r DNA, the surfactants were analyzed by TLC, FTIR and HPLC-MS. The effects of fermentation conditions on biosurfactant synthesis by strain BC1 were investigated by single factor flask shaking experiment. The properties of biosurfactants were studied. The results showed that the strain BC1 was Pseudomonas aeruginosaformis, and the main component of its surfactant was rhamnolipid Rh C10C10, after fermentation for 96 h at 37 鈩,
