【摘要】:為改善克拉瑪依油田的微生物強(qiáng)化原油開(kāi)采,從克拉瑪依油田廢水中篩選出一株性能優(yōu)良的生物表面活性劑生產(chǎn)菌株BC1。根據(jù)菌株BC1的理化指標(biāo)和16S r DNA對(duì)其進(jìn)行了屬種信息鑒定,通過(guò)薄層層析(TLC)、傅里葉紅外色譜(FT-IR)及高效液相色譜/質(zhì)譜(HPLC-MS)對(duì)其產(chǎn)生物表面活性劑進(jìn)行了分析,通過(guò)單因素?fù)u瓶實(shí)驗(yàn)考察了發(fā)酵條件對(duì)菌株BC1發(fā)酵合成生物表面活性劑的影響,研究了生物表面活性劑的性能。研究結(jié)果表明,菌株BC1為銅綠假單胞菌屬(Pseudomonas aeruginosa),其產(chǎn)生物表面活性劑主要成分為單鼠李糖脂Rh C10C10;在溫度37℃、初始p H為8、初始葡萄糖濃度5 g/L的條件下持續(xù)發(fā)酵96 h后,用酸沉降法測(cè)得鼠李糖脂產(chǎn)量為0.788 g/L;菌株BC1產(chǎn)生物表面活性劑能將發(fā)酵液表面張力從72降至28.6 m N/m,對(duì)車(chē)用機(jī)油的乳化指數(shù)E24高達(dá)93%。人工油砂剝油實(shí)驗(yàn)中,發(fā)酵液、發(fā)酵離心上清液和鼠李糖脂粗品溶液對(duì)人工油砂的剝油率均達(dá)到74%以上。菌株P(guān).aeruginosa BC1產(chǎn)生物表面活性劑在微生物強(qiáng)化采油等方面具有良好的應(yīng)用潛能。
[Abstract]:In order to improve microbial enhanced crude oil production in Karamay Oilfield, a strain BC1 with excellent performance was screened from the wastewater of Karamay Oilfield. According to the physicochemical index of strain BC1 and the information of 16s r DNA, the surfactants were analyzed by TLC, FTIR and HPLC-MS. The effects of fermentation conditions on biosurfactant synthesis by strain BC1 were investigated by single factor flask shaking experiment. The properties of biosurfactants were studied. The results showed that the strain BC1 was Pseudomonas aeruginosaformis, and the main component of its surfactant was rhamnolipid Rh C10C10, after fermentation for 96 h at 37 鈩,
