基于高通量測序技術研究玉北油田上方微生物多樣性

【摘要】:精確識別活性微生物標識物是目前微生物油氣勘探的技術核心,為深入認識各典型油氣藏上方油氣微生物標識物,采用高通量測序技術研究玉北地區(qū)油氣富集帶上方的油氣微生物物種豐度、分布、群落結構等特征.結果顯示油氣點上方樣品較之背景非油氣點具有更多的特有物種,其中油氣點樣品中含199種,背景非油氣點樣品中含77種.從中共鑒定出10種主要油氣微生物,發(fā)現(xiàn)其中的3類優(yōu)勢物種(假單胞菌屬、芽孢桿菌屬和土壤桿菌屬)是利用烴類兼性生長的微生物;同時通過解析統(tǒng)計學差異值較高的生物標記物時,發(fā)現(xiàn)兩種統(tǒng)計學差異值大于5的微生物,分別為背景非油氣點上的弧菌科Vibrionaceae維氏氣單胞菌和油氣點上的方腸桿菌科Erythrobacteraceae赤桿菌.本研究可為油氣微生物勘探提供直接實驗依據(jù).
[Abstract]:Accurate identification of active microbial markers is the core of microbial oil and gas exploration technology, in order to understand the typical oil and gas reservoirs above the oil and gas microbial markers. High throughput sequencing technique was used to study the abundance and distribution of microbial species above the oil-gas enrichment zone in Yubei area. The results show that the samples above the oil and gas point have more endemic species than the background non-oil and gas spots, among which 199 species are found in the oil and gas point samples. Background there are 77 species in non-oil and gas samples. Ten main oil and gas microbes were identified and 3 dominant species (Pseudomonas) were found. Bacillus and Agrobacterium) are microbes that use hydrocarbon facultative growth; At the same time, by analyzing the biomarkers with high statistical difference, we found two kinds of microbes with statistical difference value greater than 5. They are Vibrionaceae Vickers on the background and Erythrobacteraceae gibberellae on the oil and gas sites respectively. The results of this study are as follows:. Oil and gas microbial exploration provides direct experimental basis.
