玉米秸稈低溫高效降解復(fù)合菌系GF-20的溫度和pH適應(yīng)性研究

【摘要】:【目的】研究不同條件下復(fù)合菌系的秸稈分解能力和酶活性,評價(jià)該復(fù)合菌系的功能穩(wěn)定性及適應(yīng)性,為進(jìn)一步完善該復(fù)合菌系的篩選技術(shù)及其應(yīng)用提供理論依據(jù)!痉椒ā恳詮(fù)合菌系GF-20為研究對象,在不同溫度和pH值條件下連續(xù)繼代培養(yǎng),測定所得復(fù)合菌系的濾紙分解特性、玉米秸稈降解率、發(fā)酵液pH值、纖維素酶和半纖維素酶活性,以評價(jià)復(fù)合菌系的功能穩(wěn)定性!窘Y(jié)果】在溫度4~30℃、pH 6.0~9.0時(shí),經(jīng)過15代的繼代培養(yǎng),復(fù)合菌系GF-20在培養(yǎng)的108~140h可分解濾紙纖維素,玉米秸稈降解率保持在30%以上,發(fā)酵液pH值隨發(fā)酵時(shí)間的延長趨近于中性,pH調(diào)節(jié)能力良好,纖維素酶活性為1.22~2.34 U/mL,半纖維素酶活性為12.82~14.23U/mL。【結(jié)論】復(fù)合菌系GF-20可在較大的溫度和pH范圍內(nèi)保持較高的纖維素和半纖維素酶活性,能發(fā)揮秸稈降解作用,具有良好的開發(fā)潛力和應(yīng)用前景。
