金紋細(xì)蛾Hsp90基因片段的克隆及其表達(dá)模式分析

【摘要】:【目的】對(duì)金紋細(xì)蛾Hsp90基因進(jìn)行生物信息學(xué)和表達(dá)模式分析,為深入研究金紋細(xì)蛾P(guān)rHsp90與生長(zhǎng)發(fā)育和抗逆行為的關(guān)系奠定基礎(chǔ)�!痉椒ā坷梅肿涌寺〖夹g(shù)獲得金紋細(xì)蛾Hsp90基因片段,并對(duì)其進(jìn)行生物信息學(xué)分析。同時(shí)利用RT-qPCR技術(shù)檢測(cè)Hsp90在金紋細(xì)蛾不同組織、不同生長(zhǎng)發(fā)育階段和高溫脅迫后的表達(dá)情況�!窘Y(jié)果】獲得了金紋細(xì)蛾Hsp90基因的一條長(zhǎng)度為942bp的片段序列,命名為PrHsp90,登錄號(hào)為KP671600。序列分析顯示,其對(duì)應(yīng)的氨基酸序列與其他昆蟲的相似性很高,與棉鈴蟲(ADP37710.1)、斜紋夜蛾(ADK55516.1)、煙夜蛾(ADM26742.1)等鱗翅目昆蟲的同源性在98%以上,表明Hsp90基因家族具有高度保守性。實(shí)時(shí)熒光定量結(jié)果表明,PrHsp90在金紋細(xì)蛾翅中的相對(duì)表達(dá)量顯著高于頭、胸、腹和足;金紋細(xì)蛾各個(gè)生長(zhǎng)發(fā)育階段都有PrHsp90表達(dá),但其在成蟲體內(nèi)的相對(duì)表達(dá)量顯著高于蛹期和幼蟲期。高溫脅迫下PrHsp90的表達(dá)水平隨著溫度的升高而升高,且經(jīng)過35,38,41和44℃處理后的金紋細(xì)蛾,其體內(nèi)PrHsp90相對(duì)表達(dá)量均顯著提高,約為對(duì)照處理表達(dá)量的3~15倍�!窘Y(jié)論】金紋細(xì)蛾P(guān)rHsp90的表達(dá)具有組織特異性,且在其生長(zhǎng)發(fā)育的各個(gè)階段和溫度脅迫中均可能起重要作用。
[Abstract]:[objective] to analyze the bioinformatics and expression patterns of the Hsp90 gene of S. chrysophora. [methods] Hsp90 gene fragment was obtained by molecular cloning technique and analyzed by bioinformatics. [methods] the molecular cloning technique was used to obtain the Hsp90 gene fragment of PrHsp90. [methods] the molecular cloning technique was used to obtain the Hsp90 gene fragment, and the bioinformatics analysis of the gene fragment was carried out. At the same time, the expression of Hsp90 in different tissues, different stages of growth and development and after high temperature stress was detected by RT-qPCR. [results] A sequence of Hsp90 gene length 942bp, named PrHsp90, was obtained. Login number is KP671600. Sequence analysis showed that the amino acid sequence had high similarity with other insects. The homology of the amino acid sequence with other Lepidoptera insects such as ADP37710.1, ADK55516.1, ADM26742.1 was more than 98%, and the sequence analysis showed that the amino acid sequence was similar to other insects, such as Helicoverpa armigera (ADK55516.1), Spodoptera litura (ADK55516.1) and ADM26742.1.
