肉色吸水鏈霉菌SH121接合轉(zhuǎn)移系統(tǒng)的建立

【摘要】:利用單因素試驗(yàn)探索了熱激處理、預(yù)萌發(fā)時(shí)間、供體宿主菌的類型、供受比及MgCl_2濃度對(duì)鏈霉菌SH121接合轉(zhuǎn)移效率的影響,結(jié)果顯示:在50℃熱激10min,37℃預(yù)萌發(fā)1h,以大腸桿菌DCDA/pUZ8002為供體宿主菌,在供受比為100∶1時(shí),使用添加40mmol/L MgCl_2的培養(yǎng)基M-ISP4,接合轉(zhuǎn)移效率最高可達(dá)到每受體1.03×10~(-3)個(gè)接合子。利用建立好的接合轉(zhuǎn)移方法,將整合型質(zhì)粒pSET152和基因敲除質(zhì)粒pYZ09成功地導(dǎo)入鏈霉菌SH121,證實(shí)該方法可用于后期鏈霉菌SH121活性天然產(chǎn)物的挖掘及其相關(guān)生物合成基因簇的研究工作。
