玫瑰黃鏈霉菌Men-myco-93-63產(chǎn)抗生素發(fā)酵培養(yǎng)基的優(yōu)化

【摘要】:以玫瑰黃鏈霉菌Men-myco-93-63為發(fā)酵菌株,采用Plackett-Burman試驗(yàn)設(shè)計(jì)、最陡爬坡試驗(yàn)、中心復(fù)合試驗(yàn)設(shè)計(jì)和響應(yīng)面分析法,以抗生素產(chǎn)量為響應(yīng)值,對(duì)其發(fā)酵培養(yǎng)基進(jìn)行優(yōu)化。首先,采用Plackett-Burman設(shè)計(jì)篩選出具有正顯著效應(yīng)的花生餅粉和玉米漿。其次,通過(guò)最陡爬坡試驗(yàn)逼近最大響應(yīng)區(qū)域。最后,采用中心復(fù)合試驗(yàn)對(duì)這2個(gè)因素進(jìn)行優(yōu)化。最終確定發(fā)酵的最適培養(yǎng)基含量配方為葡萄糖2%,可溶性淀粉1%,花生餅粉4.96%,玉米漿2.66%,氯化鈉0.4%,碳酸鈣0.3%,磷酸二氫鉀0.02%。優(yōu)化后發(fā)酵培養(yǎng)基抗生素產(chǎn)量為2 030.6 mg/L,比優(yōu)化前的771.9 mg/L提高了1.63倍。
[Abstract]:Using Streptomyces roseus Men-myco-93-63 as fermentation strain, the fermentation medium was optimized by Plackett-Burman test design, steepest climbing test, central complex test design and response surface analysis. Firstly, peanut cake powder and corn pulp with positive effect were selected by Plackett-Burman design. Secondly, the maximum response region is approximated by the steepest climbing test. Finally, the central composite test was used to optimize the two factors. The optimum medium for fermentation was determined as glucose 2, soluble starch 1, peanut cake powder 4.96, corn pulp 2.66, sodium chloride 0.4, calcium carbonate 0.33, potassium dihydrogen phosphate 0.02. The antibiotic yield of the optimized fermentation medium was 2 030.6 mg/L, which was 1.63 times higher than that of 771.9 mg/L before optimization.
