一株殺撲磷高效降解菌的分離、鑒定及降解條件的響應(yīng)面法優(yōu)化

【摘要】:有機(jī)磷殺蟲劑殺撲磷對防治蚧殼蟲有特效,常用于柑橘類果樹防治但其殘留有毒有害,自然界降解速度比較緩慢.本研究分離并分類鑒定了能高效降解殺撲磷的菌株,進(jìn)一步采用Box-Benhnken法設(shè)計3因素3水平的響應(yīng)面試驗優(yōu)化降解條件.結(jié)果顯示,從富集培養(yǎng)基中分離到了一株殺撲磷高效降解細(xì)菌MS1-2,經(jīng)鑒定為膝形假單胞菌(Pseudomonas geniculata);單因素選擇溫度、NaCl濃度和pH,通過檢測殺撲磷殘留量對殺撲磷降解條件進(jìn)行響應(yīng)面優(yōu)化,轉(zhuǎn)速設(shè)置為100 r/min,接種量為0.4%時,溫度和Na Cl濃度為主要影響因素,pH為次要因素,最佳降解條件為溫度29.74℃,pH 6.48,Na Cl濃度0.63%.在此條件下培養(yǎng)24 h,殺撲磷從100 mg/L降解到57.73 mg/L,降解率為42.27%,比優(yōu)化前提高了6個百分點.因此菌株MS1-2能有效降解殺撲磷,優(yōu)化降解條件后降解率有所提高.
[Abstract]:Organophosphorus insecticide has a special effect on the control of scale insects. It is often used to control citrus fruit trees, but its residue is poisonous and harmful, and the natural degradation rate is slow. In this study, we isolated and classified the strains that could efficiently degrade triaphos, and further optimized the degradation conditions by using the Box-Benhnken method to design 3 factors and 3 levels of response surface test. The results show that A highly efficient biodegradable bacterium MS1-2, was isolated from the enrichment medium. It was identified as Pseudomonas geniculata (Pseudomonas geniculata); univariate selection temperature and pH, concentration, and the response surface was optimized by detecting the residual amount of propofol. Temperature and Na Cl concentration were the main influencing factors and pH was the secondary factor when the rotational speed was set at 100r / min and inoculation volume was 0.40.The optimum degradation conditions were as follows: temperature 29.74 鈩,
