番茄黃化曲葉病毒的鑒定與群體進化分析

【摘要】:番茄黃化曲葉病(tomato yellow leaf curl disease,TYLCD)是番茄上的重要病害,可由多種雙生病毒引起。為了明確北京地區(qū)番茄黃化曲葉病的病毒種類和病毒基因組變異情況,本研究收集了51個采自北京不同區(qū)縣的病毒疑似樣品,進行了雙生病毒的檢測及分離物的基因組分析。經(jīng)雙生病毒簡并引物檢測25個樣品呈陽性;擴增獲得10個代表性分離物的全基因組,大小均為2 781bp,共含有6個開放閱讀框;經(jīng)比對,10個分離物與Tomato yellow leaf curl virus-Israel(TYLCV-IL)同源率大于99%;基于我國TYLCV分離物全基因組序列的變異分析表明,TYLCV在進化上比較保守,整個基因組上的變異是不均勻的;系統(tǒng)發(fā)育樹顯示我國TYLCV分離物在進化樹上可分為3個組群,分析了我國TYLCV群體進化情況。
[Abstract]:Tomato yellow leaf curl disease (tomato yellow leaf curl disease TYLCD) is an important disease in tomato, which can be caused by a variety of twin viruses. In order to identify the virus species and genomic variation of tomato yellow leaf curl disease in Beijing, 51 suspected virus samples from different districts and counties in Beijing were collected for the detection of the twin virus and the genomic analysis of the isolates. 25 samples were positive by the degenerate primers of Gemini virus, and the whole genome of 10 representative isolates were obtained, which were all 2781 BP in size and contained 6 open reading frames. By comparison, the homology of 10 isolates with Tomato yellow leaf curl virus-Israel (TYLCV-IL) was greater than 99, and the variation analysis of the whole genome sequence of TYLCV isolate from China showed that TYLCV was more conservative in evolution, and the variation of the whole genome was not uniform. Phylogenetic tree showed that TYLCV isolates from China could be divided into three groups on the evolutionary tree. The evolution of TYLCV population in China was analyzed.
