金小蜂bHLH轉(zhuǎn)錄因子家族成員的鑒定與分析

【摘要】:堿性螺旋-環(huán)-螺旋(basic helix-loop-helix,b HLH)基序是蛋白質(zhì)中高度保守的可以結(jié)合DNA的結(jié)構(gòu)域。含有b HLH基序的蛋白質(zhì)組成了一個(gè)轉(zhuǎn)錄因子超家族,其家族成員廣泛存在于動(dòng)、植物以及真菌等真核生物中,在細(xì)胞增殖與分化、神經(jīng)細(xì)胞產(chǎn)生和性別決定等一系列重要發(fā)育過程中發(fā)揮調(diào)控作用。b HLH基序長約60個(gè)氨基酸,含有一個(gè)堿性區(qū)域(用于結(jié)合DNA)和由一個(gè)不定長度的環(huán)狀區(qū)域連接的兩個(gè)螺旋區(qū)域(促進(jìn)二聚化)。動(dòng)物b HLH轉(zhuǎn)錄因子根據(jù)其功能特征被分成45個(gè)家族,而這45個(gè)家族中的b HLH轉(zhuǎn)錄因子又根據(jù)其自身結(jié)構(gòu)特點(diǎn)和所作用的DNA元件被分為A、B、C、D、E和F六個(gè)高階組(high-order group)。金小蜂(jewel wasp,Nasonia vitripennis)屬于膜翅目昆蟲,為寄生蜂,是農(nóng)作物害蟲的天敵。此外,金小蜂也是一種重要的昆蟲模式生物,在復(fù)雜特征分析、發(fā)育遺傳學(xué)、進(jìn)化遺傳學(xué)等研究中得到廣泛應(yīng)用。弄清金小蜂中具有哪些b HLH轉(zhuǎn)錄因子家族成員將有助于深入了解金小蜂生長發(fā)育過程以及特殊生理生化特征產(chǎn)生的機(jī)理。本研究運(yùn)用Blast搜索、多序列比對(duì)、系統(tǒng)發(fā)生分析等生物信息學(xué)手段對(duì)金小蜂基因組中編碼的b HLH轉(zhuǎn)錄因子家族成員進(jìn)行了全面的鑒定與分析,獲得了如下結(jié)果:1、從金小蜂基因組內(nèi)成功地鑒定出了48個(gè)b HLH家族成員,根據(jù)組內(nèi)進(jìn)化分析結(jié)果將這48個(gè)b HLH家族成員歸入了36個(gè)家族和6個(gè)高階組,其中A、B、C、D、E和F六個(gè)組中分別有19、12、9、1、6和1個(gè)成員。2、蛋白質(zhì)數(shù)據(jù)庫搜索顯示,在鑒定出的48個(gè)金小蜂b HLH家族成員中,有21個(gè)成員的注釋與本研究結(jié)果一致,有17個(gè)成員的注釋與本研究結(jié)果不一致,8個(gè)成員注釋為預(yù)測蛋白,2個(gè)成員沒有注釋。因此,本研究為27個(gè)金小蜂b HLH家族成員蛋白序列進(jìn)行更為準(zhǔn)確的注釋提供了有用信息。3、通過與其它昆蟲中b HLH家族成員的分布情況進(jìn)行比較,發(fā)現(xiàn)金小蜂中在Net、Myo Ra和PTFa三個(gè)家族中沒有b HLH成員,而其它十種昆蟲在這三個(gè)家族中都有一個(gè)甚至兩個(gè)成員。這些b HLH家族成員的缺失是否與金小蜂的獨(dú)特生長發(fā)育過程有關(guān)有待進(jìn)一步探討。4、運(yùn)用電子克隆方法繪制出了29個(gè)金小蜂b HLH基因的結(jié)構(gòu)圖,其中19個(gè)基因的結(jié)構(gòu)較為完整,主要是有轉(zhuǎn)錄起始位點(diǎn)但沒有poly(A)尾;剩余的10個(gè)基因的結(jié)構(gòu)是不完整的,主要是沒有轉(zhuǎn)錄起始位點(diǎn)和poly(A)尾。本研究所獲得的結(jié)果為今后進(jìn)一步研究b HLH轉(zhuǎn)錄因子在金小蜂生長發(fā)育中的調(diào)控機(jī)理提供了有用的背景資料,也為開展不同昆蟲b HLH基因結(jié)構(gòu)與功能的比較研究打下了良好基礎(chǔ)。
[Abstract]:The basic helix-loop-helixlb HLH motif is a highly conserved DNA-binding domain in proteins. Proteins containing bHLH motifs form a transcriptional factor superfamily whose members are widely found in eukaryotes, such as animals, plants and fungi, in cell proliferation and differentiation. During a series of important developmental processes such as neuronal production and sex determination, the. BHLH motif is about 60 amino acids. It contains an alkaline region (used for binding DNA) and two helical regions (promoting dimerization) connected by a ring region of indefinite length. Animal bHLH transcription factors are divided into 45 families according to their functional characteristics, and the b HLH transcription factors of these 45 families are divided into six higher order groups (high-order group).) according to their own structural characteristics and acting elements. Jewel Waspa vitripennis belongs to Hymenoptera and is a parasitoid, which is the natural enemy of crop pests. In addition, it is also an important insect model organism, which has been widely used in complex characteristic analysis, developmental genetics, evolutionary genetics and so on. It will be helpful to understand the growth and development process and the production mechanism of special physiological and biochemical characteristics of Jumblebee by knowing which members of the bHLH transcription factor family exist in the honeybee. In this study, bioinformatics methods, such as blast search, multiple sequence alignment and phylogenetic analysis, were used to identify and analyze the members of the bHLH transcription factor family encoded in the genomes of the wasp. The following results were obtained: 1, 48 members of the b-HLH family were successfully identified from the genome of the wasp, and the 48 members of the b-HLH family were classified into 36 families and 6 higher order groups according to the results of intra-group evolution analysis. The protein database search showed that 21 of the 48 identified members of the bHLH family were consistent with the results of the present study. 17 members' comments were inconsistent with the results of this study, 8 members were annotated as predictive proteins and 2 members were not annotated. Therefore, this study provides useful information for the more accurate annotation of the protein sequences of 27 members of the bHLH family, and compared with the distribution of the members of the bHLH family in other insects. It was found that there were no bHLH members in the three families of Myo Ra and PTFa, while the other ten insects had one or two members in the three families. Whether the deletion of these members of the bHLH family is related to the unique growth and development process of the wasp needs to be further explored. By using the electronic cloning method, the structural maps of 29 bHLH genes were drawn, 19 of which were relatively complete in structure. The structure of the remaining 10 genes was incomplete, with no transcriptional initiation site and poly (A) tail. The results provided useful background data for the further study of the regulation mechanism of bHLH transcription factors in the growth and development of wasp. It also laid a good foundation for the comparative study of the structure and function of different insect bHLH genes.
