阿特拉津降解酶的提取及酶活性分析

【摘要】:以阿特拉津降解菌株為試驗材料,采用冰浴超聲破壁水提法提取粗酶,用氯仿萃取培養(yǎng)液里殘余的阿特拉津,用HPLC測定阿特拉津的殘留量,在溫度、pH、粗酶液含量不同、培養(yǎng)時間不同的條件下,研究了粗酶對阿特拉津降解率的影響,以期更好地利用阿特拉津降解酶。結(jié)果表明:阿特拉津降解酶最適宜的溫度為35℃左右,最適pH 7.5;當(dāng)粗酶液含量達(dá)到50%以上時,底物降解比較完全。
[Abstract]:Using atrazine degrading strain as experimental material, the crude enzyme was extracted by ice bath ultrasonic water extraction, and the residual atrazine was extracted by chloroform. The residue of atrazine was determined by HPLC. The content of crude enzyme was different at temperature and pH. The effect of crude enzyme on the degradation rate of atrazine was studied under different culture time in order to make better use of atrazine degrading enzyme. The results showed that the optimum temperature of atrazine degrading enzyme was about 35 鈩,
