赤霉素信號轉(zhuǎn)導(dǎo)相關(guān)基因OsGID1調(diào)控水稻對褐飛虱抗性

【摘要】:赤霉素(gibberellins, GAs)是一種重要的四環(huán)雙萜類植物生長激素,在植物生長發(fā)育中發(fā)揮著重要作用。同時(shí),赤霉素介導(dǎo)的植物生長與植物防御反應(yīng)之間存在相互制約作用,從而使植物能根據(jù)環(huán)境要求在生長與防御之間做出平衡與選擇。然而,目前對于赤霉素信號途徑調(diào)控植物防御反應(yīng)的機(jī)理還了解很少。為此,我們克隆了一個(gè)水稻赤霉素信號受體基因.-OsGID1;機(jī)械損傷、褐飛虱(Nilaparvata lugens, rice brown planthopper, BPH)為害、水楊酸(salicylic acid, SA)處理可誘導(dǎo)該基因的表達(dá),但茉莉酸(jasmonic acid, JA)處理并不產(chǎn)生誘導(dǎo)效應(yīng)。過表達(dá)OsGID1導(dǎo)致水稻SA含量顯著降低、乙烯含量顯著增加;同時(shí),OsGID1負(fù)調(diào)控GA合成酶基因OsCPS、OsKAO、 OsGA20ox的轉(zhuǎn)錄水平,而正調(diào)控GA信號途徑抑制因子基因OsDELLA轉(zhuǎn)錄水平。生測結(jié)果表明,過表達(dá)OsGID1導(dǎo)致BPH雌成蟲取食與產(chǎn)卵嗜好性降低,并引起B(yǎng)PH若蟲孵化率、單雌產(chǎn)卵量均顯著下降。上述結(jié)果表明,OsGID1在誘導(dǎo)抗蟲反應(yīng)中發(fā)揮著重要作用,作用的機(jī)理可能與OsGID1影響GA、SA、H2O2以及乙烯等信號途徑有關(guān)。
[Abstract]:Gibberellins (GAs) is an important growth hormone of dicterpenoids, which plays an important role in plant growth and development. At the same time, gibberellin mediated plant growth and plant defense response exist mutual constraints, so that plants can make the balance and choice between growth and defense according to environmental requirements. However, little is known about the mechanism of gibberellin signaling pathway regulating plant defense response. Therefore, we cloned a rice gibberellin signal receptor gene. -OsGID1; mechanical injury, Nilaparvata lugens, rice brown planthopper, BPH). Salicylic acid (SA) treatment could induce the gene expression, but jasmonic acid (JA) treatment did not induce the gene expression. Overexpression of OsGID1 resulted in a significant decrease in SA content and a significant increase in ethylene content, and a negative regulation of the transcriptional level of the GA synthase genes OsCPS- OsKAOand OsGA20ox, while the positive regulation of the GA signaling pathway inhibitor gene OsDELLA. The results of bioassay showed that overexpression of OsGID1 resulted in the decrease of feeding and spawning habits of female BPH nymphs, the hatching rate of BPH nymphs, and the decrease of oviposition quantity of single female. These results suggest that OsGID1 plays an important role in the induction of insect-resistant reactions, and the mechanism may be related to the effects of OsGID1 on the signaling pathways such as H _ 2O _ 2, H _ 2O _ 2 and ethylene.
