明尼蘇達(dá)被毛孢中活躍擴(kuò)張的HmHeli1轉(zhuǎn)座元件

【摘要】:Helitron是真核生物中存在的一種以滾環(huán)方式進(jìn)行復(fù)制的轉(zhuǎn)座元件,關(guān)于其對(duì)動(dòng)植物基因組的影響已有深入分析,而在真菌中則鮮有報(bào)道。本文對(duì)大豆囊孢線蟲(chóng)的生防菌——明尼蘇達(dá)被毛孢Hirsutella minnesotensis中的Helitron轉(zhuǎn)座元件進(jìn)行了系統(tǒng)分析,發(fā)現(xiàn)該真菌中存在兩類(lèi)自主的Helitron元件HmHeli1和HmHeli2;其中HmHeli1不僅具有保守的末端序列和結(jié)構(gòu),而且編碼序列高度相似,甚至完全一致的Rep Hel轉(zhuǎn)座酶,表明其在基因組中發(fā)生了近期擴(kuò)張并依舊活躍。HmHeli1符合Helitron2元件的結(jié)構(gòu)及編碼特征:5′和3′末端分別存在保守序列"TCAG"和"TATTTT",3′末端存在富含GC的發(fā)卡結(jié)構(gòu),近末端存在不對(duì)稱(chēng)的反向重復(fù)序列;編碼的轉(zhuǎn)座酶包含鋅指、Rep和Helicase結(jié)構(gòu)域,但不包含核酸內(nèi)切酶結(jié)構(gòu)域。HmHeli1優(yōu)先同向插入基因富集區(qū),甚至功能基因內(nèi)部,插入位點(diǎn)存在一個(gè)短的富含AT的序列。HmHeli1不具主動(dòng)捕獲基因的能力,但可被動(dòng)接受其他轉(zhuǎn)座元件的插入。HmHeli1是一種結(jié)構(gòu)獨(dú)特、極具擴(kuò)張力的DNA轉(zhuǎn)座元件,不僅在明尼蘇達(dá)被毛孢基因組進(jìn)化中發(fā)揮著重要作用,并且具有開(kāi)發(fā)為通用遺傳分析工具的潛能。
[Abstract]:Helitron is a kind of transposable element in eukaryote which is copied by rolling loop. The effect of Helitron on the genome of plant and animal has been deeply analyzed, but rarely reported in fungi. In this paper, the Helitron transposition element in Hirsutella minnesotensis, Minnesota coat, which is a biocontrol bacterium of Soybean cyst nematode, was systematically analyzed. It was found that there are two kinds of independent Helitron elements HmHeli1 and HmHeli2 in this fungus, in which HmHeli1 not only has conserved terminal sequence and structure, And the highly similar, or even identical, Rep Hel transposes, This indicates that the genome has recently expanded and is still active. HmHeli1 conforms to the structure and coding characteristics of the Helitron2 element. There are conserved sequences "TCAG" and "TATTTT" at the ends of "TCAG" and "TATTTT", respectively. There are asymmetric reverse repeats at the near end. The transposase encodes zinc finger rep and Helicase domains, but does not contain nucleic acid endonuclease domain. HmHeli1 preferentially inserts into gene rich region, even within functional gene. The insertion site has a short AT-rich sequence. HmHeli1 does not have the ability to actively capture genes, but can passively accept the insertion of other transposable elements. HmHeli1 is a DNA transposable element with unique structure and high expansion force. It not only plays an important role in the evolution of Minnesota trichoderma genome, but also has the potential to be developed as a general genetic analysis tool.
