龍門山地震帶土壤細(xì)菌多樣性的研究

【摘要】:采用16S r DNA變性梯度凝膠電泳(Denaturing Gradient Gel Electrophoresis,DGGE)技術(shù),結(jié)合不同土壤樣品的理化性質(zhì)差異,分析龍門山地震帶土壤細(xì)菌多樣性。結(jié)果表明,不同的土壤樣品其理化性質(zhì)存在著明顯的差異,三種植被類型的土壤(雜木林、竹林和松樹林)中,雜木林的土壤pH值和全氮含量最高,竹林最低;而土壤堿解氮、速效磷、速效鉀和有機(jī)質(zhì)含量則是雜木林比竹林高,松樹林最低。對DGGE優(yōu)勢條帶測序分析表明,回收的11個(gè)優(yōu)勢條帶(條帶A~K)中,條帶A、C、D、F、G和H屬于變形菌門,條帶E、J和K屬于酸桿菌門,條帶B屬于放線菌門,條帶I屬于裝甲菌門,變形菌門占細(xì)菌總類群的54.55%,說明龍門山地震帶土壤優(yōu)勢菌群為變形菌門細(xì)菌。細(xì)菌群落多樣性與土壤理化性質(zhì)相關(guān)性的統(tǒng)計(jì)分析結(jié)果顯示,影響細(xì)菌群落多樣性的主要環(huán)境因子在雜木林土壤中為速效鉀(r=0.80,P0.05),竹林為有機(jī)質(zhì)(r=0.97,P0.05),松樹林則是pH值(r=0.35,P0.05),說明龍門山地震帶不同植被類型土壤中的細(xì)菌群落對土壤理化性質(zhì)的需求不同。由于地震能夠破壞土壤結(jié)構(gòu),改變土壤理化性質(zhì),從而導(dǎo)致土壤細(xì)菌群落多樣性發(fā)生改變,因此,研究龍門山地震帶土壤細(xì)菌多樣性為震后災(zāi)區(qū)重建工作和維持龍門山地震帶生態(tài)系統(tǒng)的可持續(xù)性提供了科學(xué)依據(jù)。
[Abstract]:The bacterial diversity in Longmen Mountain seismic zone was analyzed by 16s r DNA denaturing Gradient Gel gel electrophoresis (DGGE) technique and the physical and chemical properties of different soil samples. The results showed that there were obvious differences in physical and chemical properties of different soil samples. Among the three vegetation types of soil (hybrid forest, bamboo forest and pine forest), the soil pH value and total nitrogen content of hybrid forest were the highest, and that of bamboo forest was the lowest, while that of soil alkali-hydrolyzed nitrogen was the lowest. The contents of available phosphorus, available potassium and organic matter were higher in the hybrid forest than in the bamboo forest, and the lowest in the pine forest. The sequence analysis of DGGE dominant bands showed that among the 11 reclaimed dominant bands (AK), the bands ACU, D, F, G and H belonged to Proteus, bands EJ and K belonged to acidobacterium, strip B belonged to actinomycetes, and strip I belonged to armored bacteria. Proteus accounted for 54.55% of the total bacterial group, indicating that the dominant soil flora of Longmen Mountain seismic belt was Proteus. The results of statistical analysis on the correlation between bacterial community diversity and soil physical and chemical properties showed that, The main environmental factors that affect the diversity of bacterial community are available potassium 0.80 and P0.05 in the soil of hybrid forest, organic matter of 0.97 and P0.05 in bamboo forest, and pH value of 0.35 and P0.05in the pine forest, indicating that the bacterial community in different vegetation types of soil in Longmen Mountain seismic belt is physical and chemical to the soil. The nature of the demand is different. Because earthquakes can destroy soil structure and change soil physical and chemical properties, resulting in soil bacterial community diversity change, therefore, The study of soil bacterial diversity in Longmen Mountain earthquake zone provides a scientific basis for rebuilding the disaster area after the earthquake and maintaining the sustainability of the ecological system in the Longmen Mountain seismic belt.
