塊根塊莖類作物內(nèi)生固氮菌分離鑒定、系統(tǒng)發(fā)育與促生特性

【摘要】:【目的】分離鑒定塊根塊莖類作物內(nèi)生固氮菌,研究塊根塊莖類作物內(nèi)生固氮菌的系統(tǒng)發(fā)育,分析測定塊根塊莖類作物內(nèi)生固氮菌的促生特性,探討塊根塊莖類作物內(nèi)生固氮菌的種群特點及其隨寄主植物的分布特征!痉椒ā勘砻嫦緣K根塊莖樣品后采用低氮培養(yǎng)法分離內(nèi)生細(xì)菌;通過對菌株nifH的PCR擴增、測序確認(rèn)分離細(xì)菌是固氮菌;通過16S rRNA基因測序、比對初步鑒定菌株,分析菌株的系統(tǒng)發(fā)育;通過測定菌株產(chǎn)生ACC脫氨酶、植物激素IAA,拮抗病原真菌研究菌株的促生特性!窘Y(jié)果】從胡蘿卜、白蘿卜、馬鈴薯、紫甘藍(lán)、山藥、蓮藕、芋頭、紅薯、生姜、甜菜等14個塊根塊莖類作物的塊根、塊莖中共分離到內(nèi)生固氮菌219株;16S rRNA序列,這些菌株在系統(tǒng)發(fā)育上分別屬于Acinetobacter、Arthrobacter、Bacillus、Brevibacillus、Brevibacterium、Chryseobacterium、Citrobacter、Delftia、Domibacillus、Enterobacter、Fictibacillus、Flavimonas、Flavobacterium、Microbacterium、Micrococcus、Paenibacillus,Pantoea、Pseudomonas、Rahnella、Rhizobium、Sphingobacterium、Staphylococcus、Stenotrophomonas、Variovorax,共計24屬79種,顯示了塊根塊莖內(nèi)生固氮菌豐富的種群多樣性。鑒定結(jié)果顯示,219株新分離菌株中有77株系統(tǒng)發(fā)育地位屬于Bacillus屬的23個種,29株屬于Pseudomonas屬的10個種,二者合計為33種106株,分別占系統(tǒng)發(fā)育種數(shù)和新分離菌株數(shù)的41.77%和48.40%,說明Bacillus和Pseudomonas屬是新分離塊根塊莖類作物內(nèi)生固氮菌的優(yōu)勢種群。從219株新分離菌株中選取了79株代表菌株進行促生特性研究,結(jié)果顯示8.86%菌株檢測到了ACC脫氨酶活性(0.026-13.76μmolα-丁酮酸·mg~(-1)蛋白·h~(-1)),64.56%菌株檢測到了產(chǎn)IAA(0.34-28.99μg·mL~(-1)),6.33%-13.92%菌株具有拮抗病原真菌能力(抑菌率41%-63%)!窘Y(jié)論】在正常生長的塊根塊莖類作物的塊根塊莖內(nèi)棲息著大量內(nèi)生固氮菌;塊根塊莖類作物內(nèi)生固氮菌在科學(xué)分類地位上分布廣泛,達(dá)到24屬79種,顯示出巨大的生物多樣性;Bacillus和Pseudomonas是塊根塊莖類作物內(nèi)生固氮菌優(yōu)勢種群;大約10%-60%的塊根塊莖類作物內(nèi)生固氮菌顯示出產(chǎn)生ACC脫氨酶、產(chǎn)生植物激素IAA、拮抗病原真菌等促進植物生長特性,這可能是內(nèi)生固氮菌有益于植物生長的內(nèi)在原因。
[Abstract]:[objective] to isolate and identify endophytic nitrogen-fixing bacteria in tuber tubers, to study the phylogenetic development of endophytic nitrogen-fixing bacteria in tuber tubers, and to analyze the characteristics of endophytic nitrogen-fixing bacteria in tuber tubers.The population characteristics of endophytic nitrogen-fixing bacteria in tuber tubers and their distribution with host plants were studied. [methods] Endophytic bacteria were isolated by low nitrogen culture after surface disinfection of tuber samples, and PCR amplification of the strain nifH was carried out.Sequencing confirmed that the isolated bacteria were nitrogen-fixing bacteria, analyzed the phylogeny of the strains by sequencing 16s rRNA gene, analyzed the phylogeny of the strains, and produced ACC deaminase by determining the strains.Plant hormone IAA, antagonistic characteristics of pathogenic fungi.
