基于LC-MS技術的代謝組學方法研究根腫菌對擬南芥代謝的影響

【摘要】:為探尋根腫菌侵染擬南芥后期寄主代謝的生物標志物,將罹患根腫病的油菜根切塊進行組織培養(yǎng),獲得較為純化的、單一的根腫菌接種體,與擬南芥種子在1/2MS培養(yǎng)基上共培養(yǎng),利用液相色譜-質(zhì)譜聯(lián)用(LCMS)技術的非靶向代謝組學方法分析根腫菌侵染后期擬南芥根部代謝物差異。組織培養(yǎng)結(jié)果表明,根腫菌可以在愈傷組織中生長和繁殖;共培養(yǎng)結(jié)果顯示45d后擬南芥?zhèn)雀舛四[大,顯微觀察顯示根腫菌能完成侵染循環(huán);代謝組數(shù)據(jù)PCA和PLS-DA分析結(jié)果顯示試驗組(接種根腫菌)和對照組(未接種根腫菌)明顯分離;OPLSDA分析和t檢驗結(jié)果表明重要的差異代謝物包括油菜素內(nèi)酯、類黃酮、木質(zhì)素、萜類、甾體皂苷和磷脂等物質(zhì)。提示這些物質(zhì)可能在根腫菌侵染后期時擬南芥與根腫菌的相互作用中起重要作用。
[Abstract]:In order to explore the biomarkers of host metabolism in late stage of Arabidopsis thaliana infection by Rhizomegaly bacteria, the root of Brassica napus infected by root-swelling disease was cultured by tissue culture, and a single rhizomegaly inoculum was obtained by co-culture with Arabidopsis seeds on 1/2MS medium. The difference of metabolites in root of Arabidopsis thaliana in the later stage of infection was analyzed by non-target metabolomics method of liquid chromatography-mass spectrometry. The results of tissue culture showed that rhizomegaly bacteria could grow and reproduce in callus. The results of co-culture showed that the lateral root tip of Arabidopsis thaliana was enlarged after 45 days. Microscopic observation showed that rhizomegaly bacteria could complete the infection cycle. The results of PCA and PLS-DA analysis showed that the experimental group (inoculated with root-swelling bacteria) and the control group (without inoculation of root-swelling bacteria) were significantly separated by OPLSDA analysis and t-test results showed that important differential metabolites included rapesinolactone, flavonoid, lignin. Terpenoids steroidal saponins and phospholipids suggest that these substances may play an important role in the interaction between Arabidopsis thaliana and rhizomegaly in the late stage of rhizomegaly infection.
