鉛鋅尾礦區(qū)土壤有機(jī)菌肥改良后植物根際菌群的特性及多樣性研究

【摘要】:以辣椒的根際菌群為研究對象,通過純培養(yǎng)的正交試驗(yàn)對比分析有機(jī)菌肥改良前后根際菌群去除鉛鋅能力及特性的差異,同時利用Illumina Miseq平臺對根際菌群的16S rRNA基因V4區(qū)域進(jìn)行測序,分析細(xì)菌群落結(jié)構(gòu)及多樣性的變化。結(jié)果表明:施用有機(jī)菌肥提高了可培養(yǎng)根際菌群去除鉛、鋅的能力,去除率增量最大分別可達(dá)40.2%、12.7%。同時,菌群去除鉛的最佳培養(yǎng)條件發(fā)生了改變,鉛鋅濃度和培養(yǎng)基類型對鉛去除率影響的顯著性有所提高。有機(jī)菌肥對鋅去除率的影響在試驗(yàn)分析中體現(xiàn)不明顯。培養(yǎng)基種類是影響菌群去除鉛鋅的最主要因素,菌群中的原核生物較真核生物更具去除鉛鋅的優(yōu)勢。有機(jī)菌肥的應(yīng)用對細(xì)菌群落結(jié)構(gòu)產(chǎn)生了一定影響,微生物的相對豐度發(fā)生了改變,菌群多樣性得到了豐富,但硫桿菌屬Thiobacillus仍是絕對的優(yōu)勢菌。
[Abstract]:The rhizosphere microflora of capsicum was used as the research object. The differences of the ability and characteristics of rhizosphere microflora to remove lead and zinc before and after the improvement of organic fertilizer were compared and analyzed by orthogonal experiment of pure culture. At the same time, Illumina Miseq platform was used to sequence the V4 region of 16s rRNA gene in rhizosphere microflora, and the changes of bacterial community structure and diversity were analyzed. The results showed that the application of organic fertilizer improved the ability of culturable rhizosphere microflora to remove lead and zinc. The maximum increment of removal rate can reach 40.2% and 12.77.At the same time, the optimum culture conditions for the removal of lead by bacteria have been changed. The effects of the concentration of lead and zinc and the type of culture medium on the removal rate of lead and zinc were significantly increased. The effect of organic bacterial fertilizer on the removal rate of zinc was not obvious in the experiment. The type of culture medium was the most important factor affecting the removal of lead and zinc by microbial community. Compared with eukaryotes, prokaryotes have more advantages in removing lead and zinc. The application of organic fertilizer has a certain effect on the structure of bacterial communities, the relative abundance of microbes has changed, and the diversity of bacteria has been enriched. But Thiobacillus is still the dominant bacterium.
