畢赤酵母G5拮抗葡萄灰霉病機(jī)理初探

【摘要】:針對(duì)新疆紅提葡萄中分離的生防菌株——畢赤酵母G5,以葡萄灰霉菌(Botrytis cinerea)為靶標(biāo)菌,研究了G5對(duì)葡萄灰霉菌孢子及對(duì)葡萄果實(shí)內(nèi)5種關(guān)鍵酶活性的影響,初步探討了G5拮抗葡萄采后灰霉病的機(jī)理。采用離體(in vitro)實(shí)驗(yàn)與活體(in vivo)實(shí)驗(yàn)的方法對(duì)畢赤酵母菌G5拮抗葡萄灰霉病的機(jī)理進(jìn)行探究。結(jié)果發(fā)現(xiàn),畢赤酵母菌株G5對(duì)葡萄灰霉菌孢子的萌發(fā)和菌絲生長(zhǎng)均有抑制作用,對(duì)灰霉菌絲的抑制率最高為80.20%,對(duì)灰霉孢子抑制率達(dá)86.89%;兩者間存在營(yíng)養(yǎng)競(jìng)爭(zhēng)關(guān)系,有重寄生現(xiàn)象產(chǎn)生,拮抗菌自身并不分泌抗菌物質(zhì)。此外,該菌株能誘導(dǎo)果實(shí)體內(nèi)過(guò)氧化物酶、多酚氧化酶、苯丙氨酸解氨酶、幾丁質(zhì)酶、β-1,3-葡聚糖酶的酶活性,使其酶活性有顯著提高,說(shuō)明畢赤酵母G5可以有效的誘導(dǎo)果實(shí)內(nèi)抗病相關(guān)酶的活性,增強(qiáng)對(duì)灰霉病的抑制效果。畢赤酵母G5拮抗葡萄灰霉病的機(jī)理主要包括營(yíng)養(yǎng)競(jìng)爭(zhēng)、誘導(dǎo)抗病性,是否有重寄生作用還需要進(jìn)一步驗(yàn)證。
[Abstract]:The effects of G5 on the spores of Grapevine Ash and the activities of five key enzymes in grape fruit were studied by using Botrytis cinerea, a strain of Pichia pastoris G5, isolated from red extract grape in Xinjiang as the target strain. The mechanism of antagonistic effect of G5 on gray mold of grape was studied. The mechanism of antagonistic effect of Pichia pastoris G5 on gray mold of grape was studied by in vitro and in vivo vivoo experiments. Pichia pastoris strain G5 inhibited the spore germination and hyphal growth of Aspergillus grapevine, the highest inhibition rate was 80.20%, and the inhibition rate against Aspergillus grapevine spores was 86.89%. There was a nutritional competition between the two strains, and there was a phenomenon of heavy parasitism. In addition, the antagonistic bacteria could induce the activities of peroxidase, polyphenoloxidase, phenylalanine ammonia-lyase, chitinase, 尾 -1C 3-glucanase in the fruit, and the enzyme activity was significantly increased. The results showed that Pichia pastoris G5 could effectively induce the activity of disease-related enzymes in the fruit and enhance the inhibition effect on gray mold. The mechanism of antagonistic effect of Pichia pastoris G5 against gray mold of grape mainly included nutrition competition and induced disease resistance. The existence of hyperparasitism needs further verification.
