【摘要】：【目的】揭示銅綠麗金龜Anomala corpulenta Motschulsky氣味結(jié)合蛋白(odorant binding protein,OBP)基因的表達(dá)特征及其與特異信息化合物的結(jié)合特性�！痉椒ā客ㄟ^RT-q PCR測(cè)定了銅綠麗金龜AcorOBP1基因的時(shí)空表達(dá)譜;用原核表達(dá)系統(tǒng)對(duì)AcorOBP1進(jìn)行表達(dá)純化,然后利用熒光競(jìng)爭(zhēng)結(jié)合實(shí)驗(yàn)測(cè)定了AcorOBP1蛋白與56種候選化合物的結(jié)合特征。【結(jié)果】AcorOBP1基因在銅綠麗金龜卵、1齡幼蟲和成蟲等特定發(fā)育時(shí)期表達(dá),在成蟲觸角中的表達(dá)量顯著高于其他部位(P0.05)。在被測(cè)的56種化合物中,AcorOBP1與53種化合物具有結(jié)合活性,其中結(jié)合能力較強(qiáng)的為小葉女貞Ligustrum quihoui揮發(fā)物水楊酸甲酯和癸醇,解離常數(shù)分別為2.10±0.08和5.04±0.59μmol/L。【結(jié)論】銅綠麗金龜AcorOBP1結(jié)合譜廣,該蛋白可能在銅綠麗金龜對(duì)寄主植物小葉女貞的定位過程中發(fā)揮重要作用。
[Abstract]:[objective] to reveal the expression characteristics of Anomala corpulenta Motschulsky odor binding protein (OBPs) gene and its binding to specific pheromones. [methods] the temporal and spatial expression profiles of AcorOBP1 gene were determined by RT-q PCR. AcorOBP1 was purified by prokaryotic expression system, and the binding characteristics of AcorOBP1 protein to 56 candidate compounds were determined by fluorescence competitive binding assay. [results] the AcorOBP1 gene was expressed at specific developmental stages, such as the 1st instar larvae and adults of Euonymus aeruginosa. The expression of AcorOBP1 in the antennae of adults was significantly higher than that in other parts of P0.050.AcorOBP1 had binding activity with 53 compounds, especially methyl salicylate and decanol, the volatile compounds of Ligustrum quihoui. The dissociation constants were 2.10 鹵0.08 and 5.04 鹵0.59 渭 mol / L, respectively. [conclusion] the AcorOBP1 binding spectrum is wide, and this protein may play an important role in the localization of the host plant Ligustrum lucidum.
