山東地區(qū)鹽堿土花生種子際土壤微生物群落結(jié)構(gòu)的研究

【摘要】:【目的】以不同含鹽量的濱海鹽土、內(nèi)陸鹽堿土和中等肥力非鹽堿土壤為實驗對象,探討花生種子在吸水膨脹與萌發(fā)過程中,不同類型鹽堿土對種子際土壤微生物多樣性變化的影響。【方法】采集不同含鹽量的濱海鹽土、內(nèi)陸鹽堿土和中等肥力非鹽堿土壤,通過對各樣品中細菌的16S r RNA基因的V3-V4區(qū)進行PCR擴增,利用Illumina Hiseq高通量測序技術(shù)對12份V3-V4高變區(qū)PCR產(chǎn)物進行測序,并對測序數(shù)據(jù)進行生物信息學分析�!窘Y(jié)果】(1)鹽堿土壤的種子際細菌群落多樣性高于非鹽堿土壤,且以東營青坨濱海鹽土種子際土壤細菌群落多樣性較高。(2)不同類型土壤樣本微生物群落結(jié)構(gòu)在綱水平存在明顯差異。4種土壤類型種子際土壤細菌共分屬于6個菌綱,分別為Proteobacteria、Actinobacteria、Actinobacteria、Bacteroidetes、Acidobacteria和Firmicutes菌綱,并均以Proteobacteria和Actinobacteria菌綱為主要菌綱。全樣本菌落結(jié)構(gòu)分析結(jié)果表明,4種類型土壤中不同吸脹時間內(nèi)種子際微生物菌落在門、屬水平上的類型和豐度差異最為顯著(P0.05)。(3)beta多樣性分析和各樣本遺傳距離(phylogenetic distances)聚類樹圖分析表明,4個土壤類型的12個土壤樣本種子際土壤中微生物群落均可聚為2大類。【結(jié)論】土壤含鹽量越高其種子際土壤細菌群落多樣性較高。不同類型土壤樣本微生物群落結(jié)構(gòu)在綱水平存在明顯差異,以Proteobacteria和Actinobacteria菌綱為主要菌綱。種子吸脹萌發(fā)時間影響種子際微生物菌落在門、屬水平上的類型和豐度,但對相同土壤類型樣本間遺傳距離無影響。
[Abstract]:[objective] to study the water absorption expansion and germination of peanut seeds in littoral saline soil, inland saline soil and medium fertility non-salinized soil with different salt contents. Effects of different types of saline-alkali soils on the variation of microbial diversity in interseed soil. [methods] Coastal saline soils with different salt contents, inland saline soils and moderately fertile non-salinized soils were collected. The V3-V4 region of 16s r RNA gene of bacteria was amplified by PCR, and 12 PCR products of V3-V4 hypervariable region were sequenced by Illumina Hiseq high-throughput sequencing technique. The sequence data were analyzed by bioinformatics. [results] the diversity of inter-seed bacterial community in saline-alkali soil was higher than that in non-saline-alkali soil. The microbial community structure of different soil samples in Qingtuo littoral salt soil was significantly different at the class level. 4 kinds of soil types belong to 6 classes of microbes, and the soil microbial community diversity of Qingtuo littoral saline soil was higher than that of Dongying Qingtuo littoral saline soil. The soil microbial community structure of different types of soil samples was obviously different at the class level. Proteobacteriae, Actinobacteriae, Actinobacteriae, BacteroidetesAcidobacteria and Firmicutes, with Proteobacteria and Actinobacteria as the main class respectively. The analysis of genetic distance and phylogenetic distances-cluster tree map showed that the microbial communities in 12 soil samples of 4 soil types could be clustered into 2 large soils. The results showed that the diversity of microbial communities in 12 soil samples of 4 soil types was significantly different from that of P0.05 and P0.05. The genetic distance of each sample was analyzed by phylogenetic distances-cluster tree map. [conclusion] the higher the salt content in soil, the higher the diversity of soil bacterial community in seed soil. Taking Proteobacteria and Actinobacteria as the main class, the time of seed bloating and germination affected the type and abundance of inter-seed microorganism at the level of phylum, but had no effect on the genetic distance between samples of the same soil type.
