對(duì)黃瓜枯萎病具防效的海洋源芽孢桿菌Y3F的鑒定

【摘要】:【目的】對(duì)一株從連云港海域海藻樣品中獲得的拮抗細(xì)菌Y3F進(jìn)行鑒定并且研究菌株Y3F對(duì)黃瓜土傳枯萎病害的控制效果!痉椒ā繉(duì)Y3F進(jìn)行形態(tài)、生理生化特征和16S r RNA基因序列分析,采用平板對(duì)峙法測(cè)定菌株Y3F的活菌液和無(wú)菌濾液的抑菌活性,利用盆栽試驗(yàn)測(cè)定Y3F對(duì)黃瓜枯萎病害的防治效果!窘Y(jié)果】初步鑒定該菌屬于蠟樣芽孢桿菌(Bacillus cereus),用2216E培養(yǎng)基振蕩培養(yǎng)24 h的Y3F的無(wú)菌濾液對(duì)黃瓜尖孢鐮刀菌有較強(qiáng)的抑菌活性,表明菌株能分泌抑制病原菌生長(zhǎng)的活性物質(zhì)。盆栽試驗(yàn)表明,種植30 d后,浸種和灌根同時(shí)處理(JG)的防治效果達(dá)到50.46%,對(duì)黃瓜枯萎病害有明顯的控制效果,顯著提高黃瓜植株生物量,顯著降低黃瓜根際土的真菌和尖孢鐮刀菌數(shù)量,增加根際的細(xì)菌和放線菌數(shù)量!窘Y(jié)論】菌株Y3F能有效防治黃瓜枯萎病害,改善根際微生物結(jié)構(gòu),具有進(jìn)一步開發(fā)應(yīng)用的前景。
[Abstract]:[objective] to identify an antagonistic bacterium Y3F obtained from seaweed samples from Lianyungang and study the control effect of Y3F on cucumber soil-borne Fusarium wilt. [methods] the morphology of Y3F was studied. Physiological and biochemical characteristics and sequence analysis of 16s r RNA gene were used to determine the bacteriostatic activity of live and aseptic filtrates of strain Y3F. The control effect of Y3F on Fusarium wilt of cucumber was determined by pot experiment. [results] it was preliminarily identified that Y3F belongs to Bacillus cereusus. The sterile filtrate of Y3F, which was cultured in 2216E medium for 24 h, had strong bacteriostatic activity against Fusarium oxysporum. The results showed that the strain could secrete active substances to inhibit the growth of pathogen. Pot experiment showed that the control effect of seed soaking and root irrigation at the same time reached 50.46% after planting for 30 days, and the control effect on cucumber wilt disease was obvious. The biomass of cucumber plants was significantly increased, the number of fungi and Fusarium oxysporum in the rhizosphere of cucumber was significantly decreased, and the number of bacteria and actinomycetes in rhizosphere was increased. [conclusion] strain Y3F can effectively control cucumber wilt and improve the structure of rhizosphere microorganisms. It has the prospect of further development and application.
