高通量微型生物反應(yīng)器關(guān)鍵技術(shù)研究

【摘要】:隨著生物技術(shù)的迅猛發(fā)展,微生物反應(yīng)控制系統(tǒng)逐漸由簡(jiǎn)單的人工操作向自動(dòng)化、智能化發(fā)展。微型生物反應(yīng)器逐漸成為了生物反應(yīng)器未來(lái)的發(fā)展趨勢(shì),本文針對(duì)生物反應(yīng)器微型化的問(wèn)題完成了以下三部分工作:基于PLC研制了一套微型生物反應(yīng)系統(tǒng),實(shí)現(xiàn)微生物發(fā)酵過(guò)程中溫度、酸堿度(pH)、溶解氧(DO)三個(gè)參數(shù)的監(jiān)控,提高了微生物發(fā)酵產(chǎn)物產(chǎn)量。首先研究上述三個(gè)參數(shù)對(duì)微生物發(fā)酵過(guò)程的影響,在此基礎(chǔ)上提出各個(gè)參數(shù)的控制方法,設(shè)計(jì)了微型生物反應(yīng)器的硬件架構(gòu),同時(shí)完成微反應(yīng)器上、下位機(jī)程序的編制。其中上位機(jī)程序由昆侖通態(tài)組態(tài)軟件MCGS編制,下位機(jī)控制程序由STEP7編制,上、下位機(jī)采用RS485通訊。研制了一種基于熒光猝滅原理的光纖氧傳感器,用于微生物發(fā)酵過(guò)程中溶解氧濃度檢測(cè)。分析了熒光猝滅機(jī)理,確定所使用的熒光指示劑。采用聚合物包埋的方法制備了氧傳感探頭,并對(duì)其進(jìn)行性能表征。根據(jù)氧傳感探頭的熒光特性,設(shè)計(jì)了熒光強(qiáng)度檢測(cè)電路。最后以酵母發(fā)酵、大腸桿菌發(fā)酵為實(shí)驗(yàn)對(duì)象,初步驗(yàn)證了該傳感器的可行性;贚abVIEW研發(fā)了一套的水活度智能控制系統(tǒng),實(shí)現(xiàn)非水相脂肪酶催化過(guò)程水活度智能監(jiān)控。在制定了水活度智能控制系統(tǒng)硬件架構(gòu)的基礎(chǔ)上,結(jié)合各配件工作原理運(yùn)用LabVIEW設(shè)計(jì)了該控制系統(tǒng)上位機(jī)軟件程序,該程序具有參數(shù)設(shè)定、水活度檢測(cè)、模糊自適應(yīng)PID控制以及動(dòng)態(tài)數(shù)據(jù)顯示等功能。最后以脂肪酶酯化合成三羥甲基丙烷酯過(guò)程中的水活度控制為例,驗(yàn)證了水活度控制系統(tǒng)的控制精度及穩(wěn)定性等性能。
[Abstract]:With the rapid development of biotechnology, microbial reaction control system has been developed from simple manual operation to automation and intelligence.Micro bioreactor has gradually become the development trend of bioreactor in the future. In this paper, the following three parts have been done to solve the problem of miniaturization of bioreactor: a set of micro bioreactor system based on PLC has been developed.Firstly, the effect of the above three parameters on the fermentation process of microorganism was studied. Based on this, the control method of each parameter was put forward, and the hardware structure of the micro bioreactor was designed. At the same time, the programming of the lower computer on the micro reactor was completed.The upper computer program is compiled by Kunlun on-state configuration software MCGS, and the lower computer control program is compiled by STEP7. The upper and lower computer adopts RS485 communication.An optical fiber oxygen sensor based on fluorescence quenching principle was developed to detect dissolved oxygen concentration during microbial fermentation.The fluorescence quenching mechanism was analyzed and the fluorescence indicator was determined.The oxygen sensor was prepared by polymer embedding and its properties were characterized.According to the fluorescence characteristics of oxygen sensor, a fluorescence intensity detecting circuit is designed.Finally, yeast fermentation and Escherichia coli fermentation were used as experimental objects, and the feasibility of the sensor was preliminarily verified.An intelligent control system of water activity based on LabVIEW is developed to realize intelligent monitoring of water activity in non-aqueous lipase catalytic process.On the basis of establishing the hardware structure of the intelligent control system of water activity, the software program of the upper computer of the control system is designed by using LabVIEW in combination with the working principle of various parts. The program has parameter setting and water activity detection.Fuzzy adaptive PID control and dynamic data display.Finally, the control of water activity in the synthesis of trimethylolpropane ester by lipase esterification is taken as an example to verify the control accuracy and stability of the water activity control system.
