手性熒光復(fù)合微球的制備及表征

【摘要】:本論文利用丙炔胺分別和丙烯酸及Boc-L-丙氨酸反應(yīng)合成了兩種取代乙炔類單體,即手性Boc-L-丙氨酸丙炔酰胺單體1和非手性丙烯酸丙炔酰胺單體2。兩種單體在催化劑(nbd)Rh+[η6-C6H5B-(C6H5)3]作用下進(jìn)行均(共)聚合得到產(chǎn)率57%-78%,數(shù)均分子量Mn=1.8×104-2.0 x 104的三種聚合物。圓二色光譜(CD)、紫外-可見吸收光譜(UV-vis)及旋光度([α]D)測試表明手性單體的均聚物及共聚物均具有很強(qiáng)的光學(xué)活性,并且聚合物主鏈具有單手性螺旋構(gòu)象。本論文研究了溫度、側(cè)基及非手性單體對聚合物二級結(jié)構(gòu)的影響。共聚物在三氟乙酸/二氯甲烷混合溶液作用下脫掉側(cè)鏈上的叔丁氧羰基(Boc)基團(tuán)得到帶有氨基的螺旋聚合物,這種帶有特殊官能團(tuán)的螺旋聚合物將用于下一步的接枝反應(yīng)。本論文使用無機(jī)二氧化硅納米粒子為載體制備手性熒光復(fù)合微球。首先用3-氨丙基三甲氧基硅烷及丁二酸酐對二氧化硅納米粒子進(jìn)行化學(xué)改性,使二氧化硅納米粒子表面帶有羧基,并使用滴定方法研究了二氧化硅納米粒子表面的羧基含量。其次,在脫水劑及催化劑作用下,將帶有氨基的螺旋聚合物通過共價(jià)鍵作用接枝到二氧化硅納米粒子表面。通過熱失重曲線上研究了二氧化硅納米粒子表面螺旋聚合物的接枝率。最后通過蒸餾沉淀聚合方法將熒光性能的聚合物包覆在手性復(fù)合微球表面,制備了一種新型的手性熒光復(fù)合微球。本論文利用掃描電子顯微鏡及透射電子顯微鏡觀察了手性熒光復(fù)合微球的表面形貌及粒徑。紫外-可見吸收光譜及熒光光譜表明手性熒光復(fù)合微球具有一定的熒光性能。這種手性熒光復(fù)合微球?qū)⒃谏飿?biāo)記及示蹤學(xué)領(lǐng)域發(fā)揮重要作用。
[Abstract]:In this paper, two substituted acetylene monomers were synthesized by the reaction of propargmine with acrylic acid and Boc-L-alanine, respectively. That is, chiral Boc-L-alanine alanine amide monomer 1 and non-chiral acrylic acid propionamide monomer 2.The two monomers were homopolymerized under the action of catalyst nbdfen [畏 _ 6-C _ 6B-H _ 5B _ 6H _ 5H _ 3] to obtain three kinds of polymers with average molecular weight of Mn=1.8 脳 104-2.0 x 104. The optical activity of the homopolymer and copolymers of chiral monomers were determined by dichroism, UV-vis-vis and [偽] D. And the polymer chain has a single chiral helical conformation. The effects of side group and non-chiral monomer on the secondary structure of the polymer. The copolymer removed the tert-butoxycarbonyl Boco group from the side chain in the presence of trifluoroacetic acid / dichloromethane to obtain a helical polymer with amino groups. This kind of helical polymer with special functional groups will be used in the next step of grafting reaction. In this thesis, the chiral fluorescent composite microspheres were prepared by using inorganic silica nanoparticles as the carrier. First, 3-aminopropyl trimethoxysilane and. Chemical modification of silica nanoparticles by succinic anhydride, The content of carboxyl group on the surface of silica nanoparticles was studied by titration. Secondly, under the action of dehydrating agent and catalyst, the content of carboxyl group on the surface of silica nanoparticles was studied. The helical polymers with amino groups were grafted onto the surface of silica nanoparticles by covalent bonding. The grafting rate of helical polymers on silica nanoparticles was studied by thermogravimetric curves. The polymer with fluorescent properties is coated on the surface of chiral composite microspheres by the method of deposition polymerization. A novel chiral fluorescent composite microsphere was prepared. In this paper, the surface morphology and particle size of chiral fluorescent composite microspheres were observed by scanning electron microscopy and transmission electron microscope. The spectra show that the chiral fluorescent composite microspheres have some fluorescence properties and this kind of chiral fluorescent composite microspheres will play an important role in the field of biomarker and tracer.
