【摘要】:3-羥基丙酸(3-HP)是一種重要的平臺化合物,可用于合成多種工業(yè)產(chǎn)品。利用發(fā)酵法替代化學(xué)合成法生產(chǎn)3-羥基丙酸日益受到關(guān)注,但相應(yīng)的分離技術(shù)的缺乏制約著3-羥基丙酸的大規(guī)模生產(chǎn)。絡(luò)合萃取法具有高效、處理量大、操作簡單、易于放大等優(yōu)點,在高度親水性有機物稀溶液的萃取分離中顯示了良好的應(yīng)用前景。本學(xué)位論文以有機胺類為萃取劑,開展了 3-羥基丙酸絡(luò)合萃取的研究。論文首先以三辛胺(TOA)、三癸胺和三正十二胺三種叔胺為萃取劑,比較了它們對水溶液中3-羥基丙酸的萃取能力。進(jìn)一步選取萃取能力較好的TOA為萃取劑,考察了稀釋劑種類、溶液的pH、溫度、油水相比和有機相中萃取劑體積分?jǐn)?shù)等關(guān)鍵因素對3-羥基丙酸萃取效率和分配系數(shù)的影響。在萃取溫度為293 K,相比為1:1,有機相中TOA體積分?jǐn)?shù)為30%,3-輕基丙酸初始濃度為0.330 mol/L的條件,3-羥基丙酸的萃取率達(dá)到81.6%。通過采用紅外譜光譜分析及質(zhì)量作用定律,探討了絡(luò)合萃取過程中的3-羥基丙酸萃取機理。結(jié)果表明,3-羥基丙酸可以與TOA以離子締合或氫鍵締合的形式結(jié)合。萃取平衡后的有機相中存在1:1和2:1型的酸胺萃合物,且1:1型的酸胺萃合物占主導(dǎo)地位。論文還考察了二辛胺、二癸胺和雙正十二烷基胺三種仲胺對水溶液中3-羥基丙酸的萃取效果。研究發(fā)現(xiàn),仲胺類萃取劑萃取3-羥基丙酸的能力強于烷基鏈長度相同的叔胺萃取劑。隨后探討了不同種類稀釋劑、溫度、溶液的PH、油水相比和萃取劑濃度對二癸胺萃取效率和分配系數(shù)的影響。以二癸胺和正辛醇分別為萃取劑和稀釋劑,在相比為1:1,萃取溫度為303 K的條件下,對3-羥基丙酸的分配系數(shù)為77.6,萃取率為98.8%。通過采用紅外譜光譜分析及質(zhì)量作用定律,探討了絡(luò)合萃取過程中的3-羥基丙酸萃取機理。結(jié)果表明,二癸胺通過離子締合與3-羥基丙酸結(jié)合。萃取后的有機相中,主要存在1:1和2:1兩種類型的酸胺萃合物。在研究發(fā)酵液的性質(zhì)的基礎(chǔ)上,首先采用加熱和加酸調(diào)pH相結(jié)合的方式對發(fā)酵液進(jìn)行預(yù)處理,可以高效去除其中易于產(chǎn)生乳化的蛋白質(zhì)。在此基礎(chǔ)上,將上述兩種萃取劑應(yīng)用于發(fā)酵液中3-羥基丙酸的萃取分離。研究結(jié)果表明,稀釋劑對兩種萃取劑萃取3-羥基丙酸的能力具有顯著的影響。而對于同一種稀釋劑,三辛胺對于發(fā)酵液中3-羥基丙酸的萃取能力略優(yōu)于二癸胺。利用三辛胺和正丁醇為萃取體系,通過四級萃取,3-羥基丙酸的回收率可達(dá)96.7%。
[Abstract]:3- hydroxy propionic acid (3-HP) is an important platform for the synthesis of compounds, a variety of industrial products. By fermentation method instead of chemical synthesis of 3- hydroxy propionic acid production has been paid more and more attention, but the lack of corresponding separation technology restricts the large-scale production of 3- hydroxy propionic acid. The complexation extraction method has the advantages of high efficiency, large capacity, operation simple, easy to enlarge the advantages of highly hydrophilic organic compounds in dilute solution extraction shows a good application prospect. In this thesis, using organic amine as extractant, studied 3- hydroxypropionic acid complexation extraction. The first three octylamine (TOA), three and twelve amines decylamine tri three kinds of tertiary amine as extractant, them in aqueous solution 3- hydroxypropionic acid extraction ability. Further select the better extraction capacity TOA as extraction agent, effects of diluent, solution pH, temperature, oil and water as compared to organic The key factors affecting the phase extractant volume fraction of 3- hydroxy propionic acid extraction efficiency and distribution coefficient. The extraction temperature was 293 K, compared to 1:1, TOA in the organic phase volume fraction is 30%, 3- light acid initial concentration of 0.330 mol/L, the rate of extraction of 3- hydroxy propionic acid to 81.6%. by using infrared spectrum the spectral analysis and the law of mass action, discussed 3- hydroxypropionic acid extraction mechanism of complexation extraction process. The extraction equilibrium after the organic phase of 1:1 and 2:1 type acid amine complex, and 1:1 acid amine extraction complexes dominate. By two decylamine and n-octanol as extractant and diluent respectively, in comparison to 1:1, extraction temperature conditions 303 K, the distribution coefficient of 3- hydroxy propionic acid was 77.6, the extraction rate of 98.8%. by using infrared spectra analysis and mass action law, discusses 3- hydroxypropionic acid extraction mechanism of complexation extraction process. The results showed that two decylamine by ion association with 3- hydroxy propionic acid in organic phase after extraction. The presence of 1:1 and 2:1, the main two types of acid amine complex. Based on the properties of fermentation liquid on the first use of heating and adding acid to adjust pH combination of the fermentation broth was pretreated, which can efficiently remove the easy to produce Emulsified protein. On this basis, the above two kinds of extractants used in fermentation broth of 3- hydroxy propionic acid extraction. The results show that the significant effect of diluents on the extraction of two kinds of 3- 3-hydroxypropionic ability. For the same kind of diluent, three octyl amine in fermentation solution of 3- hydroxy propionic acid extraction capacity slightly better than the two. Three using decylamine octylamine and n-butanol extraction system, through four stage extraction, 3- hydroxy propionic acid recovery rate can reach 96.7%.
