成骨細(xì)胞鈣響應(yīng)在穩(wěn)恒、脈沖流體中的實(shí)時(shí)檢測和研究(英文)

利用微流控剪切力芯片系統(tǒng)為原代成骨細(xì)胞提供準(zhǔn)確可控的流體剪切力(Fss)刺激.實(shí)時(shí)監(jiān)控成骨細(xì)胞鈣信號在穩(wěn)恒、脈沖流體作用下的變化,通過改變刺激強(qiáng)度、刺激時(shí)間、重復(fù)次數(shù)來研究成骨細(xì)胞對外部力學(xué)條件的準(zhǔn)確感知和響應(yīng).實(shí)驗(yàn)結(jié)果表明:穩(wěn)恒剪切力誘導(dǎo)的胞內(nèi)鈣響應(yīng)曲線明顯包含兩個(gè)區(qū)間:鈣峰區(qū)和平臺區(qū).鈣峰區(qū)對力學(xué)強(qiáng)度敏感,而平臺區(qū)對刺激時(shí)間敏感.在多次脈沖流體刺激下,鈣響應(yīng)隨著重復(fù)次數(shù)逐步衰減,其衰減速度與刺激間隔時(shí)間成反比.
