具有雙重治療機(jī)制的磁性納米無定型氧化鐵基藥物遞送系統(tǒng)(英文)

合成一種具有pH響應(yīng)性的聚乙二醇(PEG)修飾無定形介孔氧化鐵納米粒子(AFe-PEG).這種納米粒子可以高效負(fù)載藥物分子如阿霉素(DOX),構(gòu)成新型多功能AFe-PEG/DOX藥物遞送體系.DOX的負(fù)載率高達(dá)948 mg/g-納米粒子.在酸性溶液中,AFePEG/DOX納米粒子不僅可以有效釋放DOX,同時(shí)可以釋放Fe離子進(jìn)行Fenton反應(yīng),將H2 O2轉(zhuǎn)變成·OH自由基.體外實(shí)驗(yàn)結(jié)果表明,AFe-PEG/DOX納米粒子對(duì)HeLa細(xì)胞同時(shí)具有化療和化學(xué)動(dòng)力學(xué)療法的療效.同時(shí),由于AFe-PEG/DOX納米粒子本身的磁性,使其在外部磁場中的細(xì)胞內(nèi)化效率也得到了提高.
