低濃度納米銀對(duì)皮膚成纖維細(xì)胞和神經(jīng)內(nèi)分泌細(xì)胞增殖的影響

背景納米銀廣泛應(yīng)用于紡織品、化妝品及醫(yī)療用品等方面,可經(jīng)皮膚接觸、呼吸道、消化道等多種途徑進(jìn)入機(jī)體,增加了機(jī)體暴露于納米銀的風(fēng)險(xiǎn)。納米銀作為抗菌敷料在臨床上創(chuàng)傷表面的應(yīng)用較多,然而其潛在的應(yīng)用風(fēng)險(xiǎn)尚存在爭(zhēng)議。目的檢測(cè)低濃度納米銀暴露對(duì)大鼠乳鼠皮膚成纖維細(xì)胞和神經(jīng)內(nèi)分泌細(xì)胞增殖及細(xì)胞內(nèi)沉積的影響,為皮膚創(chuàng)傷愈合過程中納米銀的安全應(yīng)用提供理論參考。方法1.利用組織貼塊法分別取大鼠乳鼠背部和尾尖部的皮膚組織,原代培養(yǎng)皮膚成纖維細(xì)胞;常規(guī)傳代培養(yǎng)神經(jīng)內(nèi)分泌細(xì)胞;用倒置顯微鏡觀察細(xì)胞的生長(zhǎng)狀態(tài)。2.MTT法檢測(cè)不同濃度梯度的納米銀溶液孵育皮膚成纖維細(xì)胞(20-10000ng/ml)和神經(jīng)內(nèi)分泌細(xì)胞(20-5000 ng/ml)后的細(xì)胞存活率。3.選用不同終濃度的納米銀溶液分別孵育皮膚成纖維細(xì)胞和神經(jīng)內(nèi)分泌細(xì)胞,在不同的作用時(shí)間點(diǎn),倒置顯微鏡觀察活細(xì)胞形態(tài),電子顯微鏡觀察納米銀在皮膚成纖維細(xì)胞和神經(jīng)內(nèi)分泌細(xì)胞內(nèi)的沉積。4.選用終濃度為20 ng/ml、200 ng/ml、2000 ng/ml的納米銀溶液孵育皮膚成纖維細(xì)胞48 h,提取DNA,瓊脂糖凝膠電泳檢測(cè)DNA有無(wú)片段化。5.選用終濃度為20...
