載銀磷酸鈣納米線復(fù)合樹脂的制備與表征

【摘要】:隨著計算機輔助設(shè)計和計算機輔助制造技術(shù)(CAD/CAM)在口腔修復(fù)中的廣泛應(yīng)用,與CAD/CAM系統(tǒng)配套的各類可切削材料也逐漸成為研究者關(guān)注的對象,相比于陶瓷和金屬材料,可切削復(fù)合樹脂塊用于制造個性化臨時或長期使用修復(fù)體具有明顯優(yōu)勢。本論文首先通過水熱法合成了具有一維結(jié)構(gòu)的羥基磷灰石(HA)納米線,對其進(jìn)行表面改性后,得到與丙烯酸酯樹脂具有良好界面粘結(jié)的載銀HA納米線;將該HA納米線作為功能填料,添加到齒科修復(fù)常用雙酚A甲基丙烯酸縮水甘油酯(Bis-GMA)樹脂中,固化后得到一種具有抑菌效果的功能性復(fù)合樹脂,可用作可切削材料。主要工作如下:1、以油酸鈣和磷酸二氫鈉為原料,探索水熱反應(yīng)溫度和時間,在優(yōu)化反應(yīng)條件(180℃,23h)下,得到一維HA納米線;并通過多巴胺的氧化自聚合完成對該HA納米線的表面包覆改性2、 借助多巴胺的粘附性和聚乙烯吡咯烷酮的助分散作用,采用吐倫試劑(即AgNO3的氨水溶液),在HA納米線表面負(fù)載均勻分散的銀納米粒子。調(diào)節(jié)AgNO3的濃度,可控制在HA納米線表面負(fù)載的銀納米粒子含量。在不同的銀納米粒子負(fù)載量下,載銀HA納米纖維及其浸提液都表現(xiàn)出對變形鏈球菌(S.M.)的良好抑菌性;細(xì)胞毒性評價實驗顯示,當(dāng)浸提液中的銀離子濃度低于1.6ppm時,對L929成纖維細(xì)胞沒有表現(xiàn)出明顯的細(xì)胞毒性。比較抑菌實驗和細(xì)胞毒性實驗結(jié)果發(fā)現(xiàn),在兩者之間存在一定的銀離子濃度范圍(1.0-1.6ppm),使材料既具有抑菌性,又沒有明顯細(xì)胞毒性。3、選取負(fù)載合適銀納米粒子的HA納米線,以不同比例添加到Bis-GMA/TEGDMA (w:w,1:1)樹脂中,加入過氧化二苯甲酰(BPO)為引發(fā)劑,于120℃下熱固化12 h,得到復(fù)合樹脂樣品。掃描電鏡觀察和三點彎力學(xué)性能檢測顯示,當(dāng)HA納米線含量在6 wt.%-8 wt.%之間時,納米線在樹脂基體中分散良好,界面粘結(jié)性好,相比于純樹脂固化物,復(fù)合樹脂的彎曲強度提高了約20%。在純樹脂片和含不同載銀HA納米線復(fù)合樹脂片表面接種S.M.,培養(yǎng)S.M.24 h后進(jìn)行l(wèi)ive/dead染色,激光共聚焦顯微鏡下觀察顯示,純樹脂片沒有抑菌效果,而在含載銀HA納米線復(fù)合樹脂片的S.M.已經(jīng)大量死亡,表現(xiàn)出好的接觸抑菌效果。從含載銀HA納米線樹脂片的銀離子釋放檢測結(jié)果,結(jié)合前述的細(xì)胞毒性評價,可以確認(rèn),這些含載銀HA納米線復(fù)合樹脂在口腔應(yīng)用環(huán)境下,不會產(chǎn)生明顯的細(xì)胞毒性問題。
[Abstract]:With the wide application of computer-aided design (CAD) and computer-aided manufacturing (CAD/CAM) technology in oral prosthesis, various kinds of machinable materials matching with CAD/CAM system have gradually become the object of attention of researchers, compared with ceramic and metal materials. The machinable composite resin block has obvious advantages in the manufacture of personalized temporary or long-term restorations. In this paper, hydroxyapatite (HA) nanowires with one-dimensional structure were synthesized by hydrothermal method. After surface modification, silver loaded HA nanowires with good interfacial bonding with acrylate resin were obtained. The HA nanowires were added to the common bisphenol A glycidyl methacrylate (Bis-GMA) resin for dental repair. After curing, a functional composite resin with bacteriostasis effect was obtained, which can be used as cutting material. The main work is as follows: 1.
