聚脲多孔材料的簡單制備及在酶固定和手性拆分中的應用

【摘要】:以甲苯二異氰酸酯(TDI)為單體,在水與丙酮混合溶劑中通過沉淀聚合一步法制備了富含胺基的聚脲多孔材料(PPU),通過掃描電鏡和壓汞法對其表面形貌和孔結構進行了表征.PPU經(jīng)戊二醛(GA)活化后用于熒光假單胞菌脂肪酶(PFL)的固定,考察了GA活化過程中GA濃度對酶固定量及固定酶活性的影響.結果表明,PPU是一種粒子尺寸分布在30~50μm范圍的形狀不規(guī)則的多孔粒子,孔徑在2 nm~100μm之間呈連續(xù)分布.在pH=8.0的緩沖溶液中用0.17 mol/L的GA對PPU進行改性,將改性后的PPU用于PFL的固定,當酶溶液濃度為2.56 mg/m L時,得到酶的最大固定量為95.2 mg/g,固定酶的活性為375 U/mg,相對活性為76%.將此固定酶作為催化劑,用于1-苯乙醇外消旋化合物的手性拆分,并與游離酶催化的結果相比較.結果表明,固定酶的反應活性和立體選擇性都明顯優(yōu)于游離酶.通過沉淀聚合制備的聚脲多孔材料在酶固定及手性分子拆分方面具有應用前景.
[Abstract]:Using toluene diisocyanate (TDI) as monomer, Amino-rich polyurea porous materials (PPU) were prepared by precipitation polymerization in aqueous and acetone mixed solvents. The surface morphology and pore structure of PPU were characterized by scanning electron microscopy (SEM) and mercury porosimetry. PPU was activated by glutaraldehyde (GA). Immobilization of lipase from Pseudomonas fluorescens (PFL), The effects of GA concentration on enzyme fixation and enzyme activity during GA activation were investigated. The results show that nm~100 is an irregular porous particle with particle size distribution in the range of 30 ~ 50 渭 m, and its pore size is continuous between 2 nm~100 渭 m. PPU was modified with 0.17 mol / L GA in buffer solution of pH = 8.0. The modified PPU was used for immobilization of PFL. When the concentration of enzyme solution was 2.56 mg/m / L, the maximum immobilization amount was 95.2 mg / g, the activity of immobilized enzyme was 375 U / mg, and the relative activity was 76.5%. The immobilized enzyme was used as a catalyst for the chiral resolution of 1-phenylethanol racemic compounds and the results were compared with those obtained by free enzyme catalysis. The results showed that the activity and stereoselectivity of immobilized enzyme were better than that of free enzyme. Polyurea porous materials prepared by precipitation polymerization have promising applications in enzyme immobilization and chiral molecular resolution.
