云母晶面上準(zhǔn)外延生長的膠原蛋白納米線陣列的性質(zhì)研究

【摘要】:膠原蛋白纖維與同樣具有周期性結(jié)構(gòu)表面的羥磷灰石等礦物質(zhì)晶體能夠通過多種作用在介觀尺度上相互識別,從而有效地進(jìn)行規(guī)范骨骼、牙齒等器官生長的生物礦化。在本研究中,反相利用生物礦化原理,開發(fā)出一種與熱壁外延生長(Hot wall epitaxy)技術(shù)效果相似的生物大分子納米線陣列的簡單制備技術(shù),成功地將5~10μg/m L的天然I型鼠尾膠原蛋白單體溶液在云母晶體的(001)晶面上培育成取向單一且長程有序的膠原蛋白納米線陣列。原子力顯微鏡實(shí)驗(yàn)結(jié)果表明,納米線陣列隨膠原蛋白單體濃度增大而變得致密,但是單條納米線的寬度與高度較為穩(wěn)定,分別約為60.0和1.5 nm。有納米線覆蓋的云母晶面親水性好,晶面接觸角由25.8°降為9.5°;陔娮颖趁嫔⑸溲苌浜屯干潆娮语@微鏡的分析表征結(jié)果,納米線的取向應(yīng)沿云母[ˉ~110]方向,更詳實(shí)地驗(yàn)證了膠原蛋白納米線的準(zhǔn)外延生長機(jī)理。
[Abstract]:Collagen fibers and mineral crystals such as hydroxyapatite, which also have periodic structures, can be recognized on mesoscopic scales through various interactions, thus effectively biomineralizing the growth of bone, teeth and other organs. In this study, using the principle of biomineralization, a simple fabrication technique of biomolecular nanowire arrays was developed, which is similar to that of hot-wall epitaxial growth wall arrays. The natural rat tail collagen monomer solution of 5 渭 g / mL was successfully grown on the crystal plane of mica crystal to form a single and long term ordered collagen nanowire array. Atomic force microscopy (AFM) results showed that the nanowires became compact with the increase of collagen monomer concentration, but the width and height of the single nanowires were stable, about 60.0 and 1.5 nm, respectively. The mica crystal surface covered by nanowires has good hydrophilicity, and the contact angle of the crystal plane is reduced from 25.8 擄to 9.5 擄. Based on the analysis and characterization of electron backscattering diffraction and transmission electron microscope, the orientation of nanowires should be along the direction of mica. The quasi-epitaxial growth mechanism of collagen nanowires is verified in detail.
